摘要:目的观察加味黑布膏对耳郭复合痤疮模型大鼠血清血管内皮细胞生长因子(VEGF)、血小板源性生长因子(PDGF)的影响,探讨加味黑布膏外用预防痤疮瘢痕的作用机制。方法将60只SD大鼠采用随机数字表法分为空白组,模型组,阳性对照组,加味黑布膏高、中、低剂量组6组,每组10只。除空白组外,其余5组参照Kligman法建立大鼠耳郭复合痤疮模型,连续造模21 d。从实验第22天开始,于各组大鼠右耳郭造模处外涂相应药物治疗,阳性对照组外涂0.025%维A酸乳膏[5 g/(kg·d)],加味黑布膏高、中、低剂量组外涂加味黑布膏[0.08,0.04,0.02 g/(kg·d)],每日1次,连续给药4周。空白组和模型组不涂任何药物。于末次给药24 h后采用ELISA法检测大鼠血清VEGF、PDGF含量,电镜下观察大鼠皮损部位的病理改变情况。结果空白组大鼠耳郭角质层厚度较薄,毛囊皮脂腺体积正常;模型组大鼠耳郭角质层厚度明显增加,可见大量炎性细胞浸润,并有聚集,毛囊皮脂腺体积明显变大,开口处见皮脂分泌增加;各给药组上述情况均有不同程度的改善。与空白组比较,模型组大鼠血清VEGF、PDGF含量明显升高,差异均有统计学意义(P<0.01);与模型组比较,阳性对照组,加味黑布膏低、中、高剂量组大鼠血清VEGF、PDGF含量明显降低,差异均有统计学意义(P<0.01);加味黑布膏各剂量组之间比较,加味黑布膏高剂量组大鼠血清VEGF含量明显低于加味黑布膏中、低剂量组,PDGF含量明显低于加味黑布膏低剂量组,差异均有统计学意义(P<0.01)。结论加味黑布膏预防痤疮瘢痕的作用机制可能与其可调节血清中VEGF、PDGF的含量有关。
Objective To observe the effects of Jiawei Heibu Gao(加味黑布膏,JHG)on serum vascular endothelial growth factor(VEGF)and platelet derived growth factor(PDGF)of auricle compound acne model rats,and to explore the action mechanism of external application of JHG in the prevention of acne scar formation.Methods Sixty SD rats were divided into 6 groups randomly,namely blank group,model group,positive control group,JHG high,medium and low dose group,with 10 rats in each.Except the blank group,the other 5 groups established the auricle compound acne model rats by reference to Kligman method.Establishing the models for 21 consecutive days.From the 22 ndday of the experiment,corresponding medicines were applied to the right pinna of rats in each group.The positive control group was given external application of 0.025%tretinoin cream[5 g/(kg·d)],JHG high,medium,low dose group were given different doses of external application of JHG[0.08,0.04,0.02 g/(kg·d)],once a day,the medicine was administered for 4 consecutive weeks,while the blank group and model group were not given any medicines.After the last administration of 24 hours,ELISA method was used to determine the serum levels of VEGF and PDGF in rats of each group and the changes of pinna lesion in rats of each group were observed.Results In the blank group,the thickness of the auricle horny layer of the rats was thinner,the volume of hair follicle sebaceous glands was normal;in the model group,the thickness of the auricle horny layer of the rats significantly increased,a large amount of inflammatory cell infiltration was seen and aggregated,the volume of hair follicle sebaceous glands significantly enlarged,sebum secretion at the opening increased.the above conditions were improved to varying degrees in each medicine administration group.Compared with the blank group,the serum levels of VEGF and PDGF in rats of the model group all significantly increased,and the difference between the 2 groups was statistically significant(P<0.01);compared with the model group,the serum levels of VEGF and PDGF in rats of the positive control group and JHG low,medium,high dose group all significantly decreased,and the difference was statistically significant(P<0.01).Compared with JHG each dose group,the serum level of VEGF in JHG high dose group was significantly lower than that in JHG medium and low dose group,and the level of PDGF was significantly lower than that in JHG low dose group,and the difference was statistically significant(P<0.01).Conclusion The action mechanism of JHG in the prevention of acne scar formation may be related to its regulation of levels of VEGF and PDGF in serum.
