摘要:目的观察小儿开胃增食合剂对厌食症幼龄大鼠胃窦组织生长素(Ghrelin)、血管活性肠肽(VIP)及其受体表达的影响,探讨其治疗小儿厌食症的相关机制。方法将60只幼龄SD大鼠随机分为空白组、模型组、阳性对照组和小儿开胃增食合剂低、高剂量组,每组12只,采用病因模拟法建立厌食大鼠模型。各给药组给予相应药物干预,6周后采用荧光定量PCR及Western blot分别检测大鼠胃窦组织Ghrelin、VIP及其受体m RNA和蛋白表达。结果与空白组比较,模型组大鼠胃窦组织Ghrelin及其受体GHSR、VIP及其受体VPAC2m RNA和蛋白表达均显著下调(P<0.05);与模型组比较,阳性对照组、小儿开胃增食合剂高剂量组大鼠胃窦组织Ghrelin及其受体GHSR、VIP及其受体VPAC2的m RNA和蛋白表达均显著上调(P<0.05)。结论小儿开胃增食合剂通过影响模型大鼠胃窦组织Ghrelin及其受体GHSR、VIP及其受体VPAC2表达,进而改善胃的容受性舒张和紧张性收缩,调整胃的机械运动功能,发挥治疗小儿厌食症的作用。
Objective To observe the effects of Xiaoer Kaiwei Zengshi Mixture on the expressions of Ghrelin, VIP and its receptors in gastric antrum tissues of juvenile rats with anorexia;To explore the relevant mechanism for treating anorexia in children. Methods Totally sixty juvenile SD rats were randomly divided into blank group, model group, positive control group, Xiaoer Kaiwei Zengshi Mixture low-, and high-dosage groups, with 12 rats in each group. Juvenile rat model with anorexia was established by imitating the major cause of infantile anorexia. Each administration group was given relevant medicine for intervention. After 6 weeks, the mRNA and protein expressions of Ghrelin, VIP and its receptors in rat gastric antrum tissue were detected by fluorescence quantitative PCR and Western blot. Results Compared with the blank group, the mRNA and protein expressions of Ghrelin and its receptors GHSR, VIP and VPAC2 in the model group were significantly down-regulated(P<0.05);Compared with the model group, the mRNA and protein expressions of Ghrelin and GHSR, VIP and VPAC2 in the positive control group and Xiaoer Kaiwei Zengshi Mixture high-dosage groups were significantly up-regulated(P<0.05). Conclusion Xiaoer Kaiwei Zengshi Mixture can improve the gastric receptive relaxation and tension contraction, regulate the gastric mechanical function by affecting the expressions of Ghrelin and its receptors GHSR, VIP and VPAC2 in gastric antrum tissues, so as to play a role in the treatment of anorexia in children.
