摘要:目的:通过cAMP嗅觉信号转导通路探讨靶向通窍针法干预嗅觉障碍小鼠的作用机制。方法:60只BALB/c小鼠随机分为空白组、模型组、靶向通窍针刺组和药物治疗组。除空白组,其余各组制作嗅觉障碍模型,空白组和模型组不给予干预,药物治疗组给予醋酸泼尼松片4.55 mg(/kg·d),靶向通窍针刺组选取印堂、四白、迎香、列缺、合谷和风池穴针刺,各组连续治疗1个月。期间观察测定各组小鼠的埋食时间,并用ELISA试剂盒测定各组小鼠治疗前后cAMP浓度,探讨cAMP信号转导通路在靶向通窍针法干预嗅觉障碍中的作用。结果:治疗前与空白组比较,模型组cAMP浓度显著升高(P<0.05)。治疗后,靶向通窍针刺组cAMP浓度高于模型组及空白组(P<0.05),且与药物治疗组相当。与空白组比较,模型组小鼠埋食时间显著延长(P<0.01),靶向通窍针刺组小鼠埋食时间与模型比较显著减少(P<0.01),且与药物治疗组相当。结论:靶向通窍针法可改善小鼠的嗅觉障碍,其机制可能与靶向通窍针法上调嗅觉障碍信号转导通路中的cAMP浓度有关。
Objective:To explore the action mechanism of targeted Tongqiao acupuncture in intervening olfactory disorder(OD)through the cAMP olfactory signal transduction pathway in mice.Methods:60 BALB/c mice were randomly divided into the blank group,the model group,the targeted Tongqiao acupuncture group,and the medication group.OD model was prepared in all groups except the blank group.The blank group and the model group were not given any intervention.The medication group was treated with Prednisone Acetate Tablet at the dose of 4.55mg(/kg·d);the targeted Tongqiao acupuncture group was treated with needling Yintang(EX-HN3),Sibai(ST2),Yingxiang(LI20),Lieque(LU7),Hegu(LI4)and Fengchi(GB20);each group was treated continuously for 1 month.During this period,the time of finding the buried food was observed in mice,the ELISA kit was used to measure the cAMP concentration before and after the treatment,the role of cAMP signal transduction pathway was explored in the therapy of targeted Tongqiao acupuncture intervening OD.Results:Compared with that in the blank group,the concentration of cAMP was significantly increased in the model group before the treatment(P<0.05).After the treatment,the concentration of cAMP was higher in the targeted Tongqiao acupuncture group than that in the model group and the blank group(P<0.05),which was similar to that in the medication group.Compared with that in the blank group,the time of finding buried food was significantly prolonged in the model group(P<0.01);the time of finding buried food was significantly shortened in the targeted Tongqiao acupuncture group compared with that in the model group(P<0.01),which was similar to that in the medication group.Conclusion:Targeted Tongqiao acupuncture can improve OD in mice,and the mechanism may be related to the up-regulation of cAMP concentration in the signal transduction pathway of OD.
