摘要:目的研究煎煮形式及配伍比例对党参黄芪复方免疫调节作用的影响。方法按照体重将昆明种小鼠随机分为9组:正常组、模型组、对照组、党参组、黄芪组、3个不同配比合煎组(党参与黄芪,1∶1,1∶2,2∶1)及单煎合并组(党参与黄芪,1∶1),每组10只。用环磷酰胺40 mg·kg^(-1)制备小鼠免疫低下模型。6个实验组分别灌胃受试样品14.3 g·kg^(-1),对照组灌胃参芪颗粒5 g·kg^(-1);正常组和模型组灌胃等量0.9%Na Cl,每天给药1次,连续15 d。以称重计算小鼠脏器指数,以腹腔注射5%鸡红细胞悬液(CRBC)测定腹腔巨噬细胞吞噬百分率及吞噬指数,以50%二硝基氯苯(DNCB)丙酮溶液致迟发型超敏反应测定OD值和血清溶血素OD值,以尾静脉注射印度墨汁测定网状内皮系统吞噬指数及吞噬系数。结果与模型组的胸腺指数、脾指数分别为(0.64±0.24),(1.83±0.89 g·kg-1)比较,党参组、黄芪组、单煎合并组、3个不同配比合煎组(1∶1,1∶2,2∶1)的胸腺指数(g·kg^(-1))分别为1.23±0.73,1.42±0.43,1.47±0.41,1.97±1.05,1.82±0.99,1.49±0.59和脾指数(g·kg^(-1))分别为2.09±2.61,2.38±1.43,2.40±1.39,2.43±1.44,2.58±1.05,2.36±0.89,差异有统计学意义(P<0.05或P<0.01)。与模型组的吞噬百分率为(22.45±3.55)%和网状内皮系统吞噬指数为0.02±0.01比较,这6个实验组吞噬百分率(%)分别为30.41±7.28,39.96±10.45,42.23±7.43,44.83±7.95,43.75±10.60,40.43±8.91;而吞噬指数分别为0.04±0.03,0.04±0.04,0.05±0.02,0.07±0.04,0.06±0.03,0.04±0.02,差异有统计学意义(均P<0.01)。与模型组(迟发型超敏反应OD值0.24±0.05,血清溶血素OD值0.28±0.11)比较,这6个实验组的迟发型超敏反应OD值分别为0.37±0.16,0.40±0.12,0.42±0.17,0.50±0.25,0.46±0.17,0.44±0.20;而血清溶血素OD值分别为0.58±0.08,0.64±0.12,0.48±0.14,0.68±0.08,0.54±0.18,0.64±0.06,差异有统计学意义(均P<0.05或P<0.01)。2个配比合煎组(1∶1,1∶2)作用效果均优于党参组和黄芪组。结论党参与黄芪两味中药配伍使用时的免疫调节活性均强于两药单独使用,有显著的协同作用。
Objective To study the effect of decoction forms and compatibility proportion of Codonopsis and Astragalus compound on immuno- modulatory. Methods The kunming mice were randomly divided into nine groups : normal, model, control, test - Codonopsis, test - Astraga- lus, test- merged with individual decocted, test- Codonopsis and Astragalus ( 1 : 1 ), ( 1 : 2 ), ( 2 : 1 ) , each group had ten rats. Making low- immunity model mice by cyclophosphamide( CTX 40 mg· kg-1 ), The normal and model were administrated the same quantity of normal saline, the control were administrated the Shenqi particle (5 g·kg^-1 ), test - Codonopsis, test - Astragalus, test - merged with individual decocted, test - Codonopsis and Astragalus ( 1 : 1, 1 : 2, 2:1 ) were administrated the corresponding equal doses test agent( 14. 3 g·kg^-1 )for 15 days. The visceral index was counted by weighted. The peritoneal macrophage phagocytic count by intraperitoneal injection 5% chicken red blood -cell(CRBC) were meaeured( OD value) . The serum hemolysin OD through delayed type hypersensitivity maded by 50% dinitrochloro- benzene (DNCB). The reticuloendothelial system phagocytic count were meaeured by caudal vein injection carbon inkl. Results Compared with model group ( thymus index 0. 64 ± 0. 24 g·kg^-1, spleen index 1.83 ± 0. 89 g·kg^-1), the thymus index ( g·kg^-1 ) in test - Codonopsis, test - Astragalus, test - merged with individual decocted, test - Codonopsis and Astragalus ( 1: 1, 1: 2, 2:1 ) were 1.23 ± 0. 73, 1.42 2 0. 43,1.47 ± 0. 41, 1.97 ± 1.05, 1.82± 0. 99, 1.49 ± 0. 59, all can varying degrees improve with siginificantly ( P 〈 0.05 or P 〈 0.01 ) and spleen index( g·kg^-1) in above groups were 2. 09 ± 2.61, 2. 38±1.43, 2. 40± 1.39, 2. 43±1.44, 2.58± 1.05, 2. 36 ± 0. 89 with siginificantly ( P 〈 0.05 or P 〈 0. 01 ). Compared with model group, the peritoneal macrophage phagocytic count ( % ) in above groups were 30. 41 ± 7.28, 39. 96± 10. 45, 42.23 ± 7.43, 44. 83 ± 7.95, 43.75 ± 10. 60, 40. 43 ± 8.91 and the reticuloendothelial system phagocytic countin above groups were 0. 04±0. 03, 0. 04 ± 0. 04, 0. 06 ± 0.02, 0. 07 ± 0. 04, 0. 06 ± 0. 03, 0. 04 ± 0. 02 with siginificantly ( P 〈 0. 05 or P 〈 0. 01 ). Compared with model group ( OD value 0. 24 ±0.05,0.28±0.11 ), the restrain delayed type hypersensitivityin above groups were 0. 37 ±0. 16, 0. 40 ±0. 12, 0. 42 ±0. 17, 0.50 20. 25,0.46 ±0. 17, 0.44 20. 20 in immunosuppressed mice induced by CTX and serum hemolysin OD in above groups were 0. 58±0. 08, 0. 64± 0. 11, 0. 48 ± 0. 14, 0.68±0.08, 0. 54±0. 18, 0. 64±0. 06 with siginificantly ( P 〈 0. 05 or P 〈 0. 01 ). Comparatively speaking, the effect of test - Codonopsis and Astragalus ( 1 : 1 ) and (1:2) were overmatch the groups that decocted individual. Conclusion The compatibility and synergy of Codonopsis and Astragalus can enhance their immunoregulatory activity.
