摘要:目的观察芪蛭皱肺颗粒对慢性阻塞性肺疾病(COPD)模型大鼠肺组织和血清中超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)水平及肺组织病理结构的影响,探讨其阻抑COPD的效应及机制。方法 Wistar大鼠随机分为空白组、模型组、阳性对照组与芪蛭大、中、小剂量组。采用气管内滴注脂多糖(LPS)加烟熏法复制COPD大鼠模型,造模后第15日开始药物干预,第43日分别检测血清、肺组织SOD、MDA、NO水平及肺组织病理形态的变化。结果与空白组比较,模型组大鼠肺组织与血清中SOD、NO含量均降低(P<0.05),MDA含量明显升高(P<0.05)。与模型组比较,芪蛭大、中剂量组肺组织与血清中SOD含量均显著升高(P<0.05),芪蛭大、中剂量组肺组织中MDA含量均显著降低(P<0.05),芪蛭大、小剂量组血清中MDA含量均显著降低(P<0.05),芪蛭大、小剂量组肺组织中NO含量明显升高(P<0.05),芪蛭大、中剂量组血清中NO含量明显升高(P<0.05)。各治疗组大鼠肺组织形态学改变较模型组减轻。结论芪蛭皱肺颗粒对COPD大鼠肺组织具有保护作用,提高抗氧化能力、调节NO代谢可能是其干预COPD发生、发展的机制之一。
Objective To observe the effects of Qizhi Zhoufei Granules on SOD,MDA,NO of lung tissue and serum and the pathology structure of lung tissue of chronic obstructive pulmonary disease(COPD) model rats,and explore its mechanism of preventing and treating COPD.Methods Wistar rats were randomly divided into blank group,model group,positive control group,Qizhi low-dose group,Qizhi medium-dose group and Qizhi high-dose group.COPD model was established by smoking and infusing lipopolysaccharide(LPS).After 15 days,the rats were given the drugs by gavage.On the 43rd day,the change of SOD,MDA,NO in serum and lung tissue and pathology of lung tissues were measured.Results Compared with the blank group,the levels of SOD and NO of lung tissue and serum were lowered in model group(P 0.05),and the level of MDA were raised(P 0.05).Compared with the model group,SOD level of lung tissue and serum in Qizhi high-and medium-dose group were raised(P 0.05),and MDA level of lung tissue in Qizhi high-and low-dose group were obviously lowered(P 0.05),the NO level of lung tissue in Qizhi high-and low-dose group were raised(P 0.05),the NO level of serum in Qizhi highand medium-dose group were raised(P 0.05).In all treated groups,the morphology of lung tissue was alleviated compared with the model group.Conclusion Qizhi Zhoufei Granules have protective effect on lung tissue of COPD rats.It can raise the oxidation resistance and adjust the NO metabolism,which may be one of the mechanisms of intervening occurrence and development of COPD.
