摘要:目的:观察痛泻要方对溃疡性结肠炎(UC)模型大鼠结肠黏膜PPAR-γ蛋白和基因表达的影响,探讨其作用机制。方法:将实验动物分为6组,采用TNBS/乙醇灌肠法造模,肉眼观察结肠大体形态损伤并进行评分,以大鼠结肠黏膜PPAR-γ为观察指标,采用RT-PCR和免疫组化法检测PPAR-γ蛋白和基因表达水平。结果:肉眼观察模型组大鼠结肠组织黏膜层可见炎症和溃疡形成,与空白组比较差异有统计学意义(P<0.05),证实模型成功。模型组大鼠结肠组织PPAR-γ基因和蛋白的表达量均低于空白组,差异有统计学意义(P<0.01);治疗后,痛泻要方高剂量组、中剂量组PPAR-γ基因和蛋白的表达量均较模型组升高,差异有统计学意义(P<0.05,P<0.01)。结论:痛泻要方对TNBS/乙醇法UC大鼠模型结肠黏膜PPAR-γ基因和蛋白的表达量有上调作用,提示痛泻要方治疗UC的作用可能与PPAR-γ/NF-κB信号转导途径被激活有关。
Objective: To observe the influence of Tong Xie Yao Fang on the PPAR-γ mRNA and protein expressions of colonic mucosa in rats with experimental ulcerative colitis(UC),and identify its underlying mechanism of action.Methods: Dividing the experimental animals into 6 groups,we prepared the models by means of the TNBS/ethanol enema,observed their morphological injury and damaged with the naked eye,and detected PPAR-γ gene and protein expressions by means of RTPCR and immunohistochemical with PPAR-γ being the observation targets.Results: Inflammation and ulceration were with the naked eye observed on the colonic mucous membrane of rats in the model group,and compared with the blank group,and the models were proved successful(P0.05).The relative expression amount of PPAR-γ gene and protein of colonic tissues of rats in the model group were decreased than that of the blank group,and the discrepancy was statistically significant(P0.01);after the treatment,compared with the model group,the relative expression amount of PPAR-γ gene and protein in the Tong Xie Yao Fang high-and moderate-dose groups were increased,and the discrepancy was statistically significant(P0.05,P0.01).Conclusion: The Tong Xie Yao Fang can promote the relative expression amount of PPAR-γ gene and protein of colonic tissues of rats with UC,which suggests the therapeutic effects of the Tong Xie Yao Fang is probably related to the activation of PPAR-γ/NF-κB signal transduction pathway.
