摘要:目的观察红芪多糖(HPS)对糖尿病周围神经病变ob/ob小鼠运动神经传导速度(MNCV)和神经核转录因子-κB(NF-κB)与白细胞介素-1β(IL-1β)表达的影响。方法按照体重将ob/ob小鼠随机分为5组:模型组、对照组(α硫辛酸30 mg·kg-1)和低、中、高3个剂量实验组(50,100,200 mg·kg-1HPS),每组10只;另设C57BL/6野生型小鼠10只作为正常组,均灌胃给药,1次/天,连续8周。末次给药后,用生理记录仪测定小鼠MNCV;用免疫印迹法和逆转录-聚合酶链反应法检测小鼠的NF-κB、IL-1β的蛋白与mRNA表达情况。结果给药8周后,低、中、高3个剂量实验组、对照组和模型组的MNCV分别为(40. 35±2. 21),(40. 34±2. 23),(43. 74±3. 14),(42. 70±3. 49)和(40. 29±2. 90) m·s-1,高剂量实验组和对照组与模型组比较,差异均有统计学意义(P <0. 01或P <0. 05)。中、高2个剂量实验组、对照组和模型组的NF-κB蛋白表达灰度值分别为0. 35±0. 02,0. 31±0. 03,0. 29±0. 03,1. 27±0. 09;这4组的IL-1β蛋白表达灰度值分别为0. 28±0. 03,0. 25±0. 03,0. 23±0. 03,1. 35±0. 01,中、高2个剂量实验组与模型组比较,差异均有统计学意义(均P <0. 01)。基因结果的趋势与蛋白一致。结论 HPS能提高小鼠的MNCV,降低NF-κB、IL-1β蛋白与mRNA的表达水平,减轻其炎症反应状态。
Objective To observe the effect of Hedysarum polybotrys saccharide( HPS) on the motor nerve conduction velocity( MNCV) and the expression of nuclear factor Kappa B( NF-κB) and interleukin IL-1 beta( IL-1β) in diabetic peripheral neuropathy ob/ob mice.Methods The ob/ob mice were randomly divided into 5 groups by weight: model group,control group( alpha lipoic acid 30 mg·kg^-1),and experimental-L,experimental-M,experimental-H groups( 50,100,200 mg·kg^-1 HPS),each group had 10 mice;and 10 wild type C57 BL/6 mice as normal group.The rats were given orally once a day for 8 weeks.After the last administration,the MNCV was determined by electrophysiolograph.The expression of NF-κB and IL-1β protein and mRNA were detected by Western blotting and reverse transcription polymerase chain reaction.Results Eight weeks after treatment,the MNCV in experimental-L,experimental-M and experimental-H groups,control group and model group were( 40.35±2.21),( 40.34±2.23),( 43.74±3.14),( 42.70±3.49) and( 40.29±2.90) m·s^-1,respectively.Comparison between experimental-H and control group with model group,the difference was significant( P < 0.01 or P < 0.05).The gray level of NF-κB protein expression in experimental-M and experimental-H groups,control group and model group were 0.35±0.02,0.31±0.03,0.29±0.03,1.27±0.09,respectively.The gray values of IL-1β protein expression in the four groups were0.28±0.03,0.25±0.03,0.23±0.03,1.35±0.01,respectively.Comparison between experimental-M,experimental-H groups with model group,the difference of the factors were significant( all P < 0.01).The trend of gene results was consistent with that of protein.Conclusion HPS can increase the MNCV.HPS can decrease the expression of NF-κB,IL-1β protein and mRNA in the nerve tissue of ob/ob mice.
