摘要:【目的】优选牡丹花粉口含片的处方和制备工艺,并对其抗氧化活性进行评价.【方法】以破壁的牡丹花粉为原料,采用湿法制粒压片,通过考察口含片的外观、口感、硬度、崩解时限,确定牡丹花粉口含片的最佳处方和制备工艺,并通过测定牡丹花粉口含片模拟体内消化产物对DPPH·和ABTS·的清除能力,评价其抗氧化能力.【结果】优选得到的处方和工艺为牡丹花粉22%、甘露醇-乳糖(1∶2)67%、阿斯巴甜-柠檬酸(1∶1.5)11%,混合均匀,用70%乙醇湿法制粒,烘干,整粒,加入1.5%硬脂酸镁,压片,即得.牡丹花粉口含片经模拟人体消化后,唾液、胃液和肠液的消化产物均有一定的清除DPPH·和ABTS·的能力,累加消化液的消化产物对DPPH·、ABTS·的IC50分别为(4.722±0.112)(0.540±0.048)mg/mL.【结论】牡丹花粉口含片处方合理,工艺稳定可行,具有较好的抗氧化活性.
【Objective】To optimize the prescription and preparation process of peony pollen buccal tablet,and evaluate its anti-oxidant activity.【Method】The wall-broken peony pollen was used as the raw material to prepare the buccal tablet by wet granulation process,and the appearance,taste,hardness and disintegrating time were used as indicators to optimize prescription and preparation process.The antioxidant activity of the tablet was evaluated by measuring the DPPH and ABTS free radical scavenging ability of simulated digestion products.【Result】The optimal formulation was 22%peony pollen,67%mannitol-lactose(1∶2)and 11%aspartame-citric acid(1∶1.5),and then buccal tablet was prepared according to the prescription with 70%ethanol granulation and 1.5%magnesium stearate.Under the condition of simulated digestion in vitro,the samples simulated saliva,gastric juice and intestinal fluid digestion had certainly DPPH and ABTS free radical scavenging activity.The half elimination ratio(IC50)of the accumulated digestion sample on DPPH and ABTS free radicals was(4.722±0.112)mg/mL and(0.540±0.048)mg/mL respectively.【Conclusion】The optimum prescription was reasonable,and the optimal preparation process was stable and feasible.The peony pollen buccal tablet had significant antioxidant activity.
