摘要:胃癌是消化系统最常见的恶性肿瘤之一,由于其发病隐匿,多数患者发现时已处于晚期。由于胃癌是一种多功能疾病,其发病机制目前尚不完全明确。多种信号通路的激活对胃癌的进展至关重要,而PI3K/Akt/mTOR信号通路备受关注。基因突变及表观遗传修饰可引起该通路失调从而可导致胃癌发生,且该通路参与胃癌的增殖、侵袭、转移及耐药等多种病理过程。因此,本文将PI3K/Akt/mTOR信号通路在GC发生发展以及基于该通路的抑制剂在GC中的作用机制作一总结概括,以期为胃癌的诊断及治疗提供依据。
Gastric cancer(GC)is one of the most common malignant tumors of the digestive system,and most patients are found at an advanced stage due to its insidious onset.Since GC is a multifunctional disease,its pathogenesis is not fully understood.Studies have shown that the activation of multiple signaling pathways is crucial for the progression of GC,and the phosphoinositide 3-kinase(PI3K)/protein kinase B(AKT)/mammalian target of rapamycin(mTOR)signaling pathway has attracted much attention.Gene mutations and epigenetic modifications result in the dysfunction of the PI3K/Akt/mTOR signaling pathway,leading to the occurrence of GC.It is also involved in various pathological processes,such as proliferation,invasion,metastasis and drug resistance of GC.Therefore,this article summarized the occurrence and development of PI3K/Akt/mTOR signaling pathway in GC and the mechanism of action of inhibitors based on this pathway in GC,thus providing a basis for the diagnosis and treatment of GC.
