摘要:目的观察参苓白术散对抗生素诱导的肠道菌群失调幼鼠肺部免疫炎症反应的影响。方法60只雄性Balb/c幼鼠随机抽取10只作为空白组,其余幼鼠采用混合抗生素灌胃诱导肠道菌群失调,连续21 d,末次灌胃后12 h,按随机数字表法将造模幼鼠分为模型组、阳性对照组(双歧杆菌四联活菌片0.68 g/kg)及参苓白术散高、中、低剂量组(29.12、14.56、7.28 g/kg),每组10只,分别灌胃相应药液,空白组和模型组灌胃生理盐水,每日1次,连续7 d。末次灌胃后收集幼鼠粪便,采用16S rDNA测序分析肠道菌群。除空白组外,其余各组经口咽吸入脂多糖诱导肺部炎症反应,7 d后HE染色观察肺组织病理变化,并进行病理学评分,ELISA检测支气管肺泡灌洗液白细胞介素-10(IL-10)、干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)含量及血清免疫球蛋白(Ig)G、IgM、IgA含量。结果与空白组比较,模型组幼鼠肠道菌群Shannon指数明显降低(P<0.01),变形菌门丰度明显增加(P<0.01),拟杆菌门、厚壁菌门丰度明显减少(P<0.01);与模型组比较,参苓白术散各剂量组幼鼠肠道菌群Shannon指数明显升高(P<0.01),变形菌门丰度明显减少(P<0.01),拟杆菌门、厚壁菌门丰度明显增加(P<0.01),参苓白术散高、低剂量组幼鼠肠道疣微菌门丰度增加(P<0.05,P<0.01)。HE染色显示,与空白组比较,模型组幼鼠肺组织损伤严重,病理评分明显升高(P<0.01);与模型组比较,参苓白术散各剂量组幼鼠肺组织损伤有所减轻,病理评分明显降低(P<0.05,P<0.01)。ELISA结果显示,与空白组比较,模型组幼鼠支气管肺泡灌洗液IL-10含量明显减少(P<0.01),IFN-γ、TNF-α含量明显增加(P<0.01),血清IgM含量明显增加(P<0.01);与模型组比较,参苓白术散高剂量组幼鼠支气管肺泡灌洗液IL-10含量明显增加(P<0.05,P<0.01),IFN-γ、TNF-α含量明显减少(P<0.01),血清IgM含量明显减少(P<0.01)。结论参苓白术散可调节肠道菌群失调幼鼠菌群结构,改善其肺部免疫炎症反应。
Objective To observe the effects of Shenling Baizhu Powder on the immune inflammatory response in the lungs of juvenile mice with intestinal flora imbalance induced by antibiotics.Methods Ten of the 60 male Balb/c juvenile mice were randomly selected as the blank group.The remaining fifty juvenile mice were treated with mixed antibiotics by gavage for 21 consecutive days.After the last treatment for 12 hours,the juvenile mice were divided into model group,positive control group(bifidobacterium quadruple viable bacteria tablets 0.68 g/kg),and Shenling Baizhu Powder high,medium-and low-dosage groups(29.12,14.56,7.28 g/kg)according to random number table method,with 10 mice in each group.The administration groups were given corresponding drugs by gavage,the blank group and the model group were given normal saline by gavage,once a day,for 7 consecutive days.After the last gavage,feces were collected,the intestinal flora was analyzed by 16S rDNA sequencing.Except for the blank group,the other groups inhaled lipopolysaccharide(LPS)through oropharynx to induce pulmonary inflammatory response.After 7 days of inflammatory response,the morphology and structure of lung tissue were observed by HE staining and pathological score was performed,ELISA was used to detect the contents of interleukin-10(IL-10),interferonγ(IFNγ),tumor necrosis factorα(TNF-α)in bronchoalveolar lavage fluid and immunoglobulin(Ig)G,IgM and IgA in serum.Results Compared with the blank group,the intestinal flora Shannon index of juvenile mice in the model group decreased significantly(P<0.01),the abundance of Proteobacteria increased significantly(P<0.01),the abundance of Bacteroidetes and Firmicutes decreased significantly(P<0.01).Compared with the model group,the intestinal flora Shannon index of juvenile mice in each dosage of Shenling Baizhu Powder groups significantly increased(P<0.01),the abundance of Proteobacteria decreased significantly(P<0.01),the abundance of Bacteroidetes and Firmicutes increased significantly(P<0.01),the abundance of Verrucomicrobiota in Shenling Baizhu Powder high-and low-dosage groups increased(P<0.05,P<0.01).HE staining showed that compared with the blank group,the lung tissue injury of juvenile mice in the model group was serious and the pathological score increased significantly(P<0.01).Compared with the model group,the lung tissue injury and pathological score of juvenile mice in each dosage of Shenling Baizhu Powder groups decreased significantly(P<0.05,P<0.01).ELISA result showed that compared with the blank group,the content of IL-10 in bronchoalveolar lavage fluid of juvenile mice in the model group reduced significantly(P<0.01),the contents of IFN-γand TNF-αincreased significantly(P<0.01),the content of IgM in serum increased significantly(P<0.01).Compared with the model group,the content of IL-10 in bronchoalveolar lavage fluid of juvenile mice in Shenling Baizhu Powder high dosage group increased significantly(P<0.05,P<0.01),the contents of IFN-γand TNF-αreduced significantly(P<0.01),the content of IgM in serum reduced significantly(P<0.01).Conclusion Shenling Baizhu Powder can regulate the flora structure of juvenile mice with intestinal flora imbalance and improve the immune and inflammatory response of lungs.
