摘要:目的:探讨镇肝熄风汤对自发性高血压大鼠(SHR)肠道菌群的影响。方法:50只14 w龄雄性SHR大鼠随机分为模型对照组、贝那普利0.9 mg/kg组、镇肝熄风汤15、30、60 g/kg组,10只WKY大鼠设为正常对照组。每日分别给予相应药物或蒸馏水灌胃。干预8 w后测定各组大鼠血压,取大鼠盲肠、结肠粪便,进行16SV3-V4区扩增子信息采集与分析。结果:与正常对照组比较,模型对照组大鼠收缩压、舒张压、平均动脉压明显升高(P<0.05);盲肠Turicibacter属丰度明显升高,丁酸梭菌属丰度明显降低(P<0.05);结肠乳酸杆菌属丰度明显升高,粪球菌属、瘤胃球菌属丰度明显降低(P<0.05);与模型对照组比较,各给药组收缩压、舒张压和平均动脉压明显降低(P<0.05);各给药组变形菌门丰度减少,盲肠乳酸产生菌Turicibacter属相对丰度下降,丁酸产生菌丁酸梭菌属相对丰度明显上升(P<0.05);结肠乳酸产生菌乳酸杆菌属相对丰度下降,丁酸产生菌粪球菌属和瘤胃球菌属相对丰度上升(P<0.05);Alpha多样性显示,各干预组多样性上升;Beta多样性显示,各干预组分布较模型对照组集中。结论:镇肝熄风汤可以降低SHR血压,其机制可能与降低肠道乳酸产生菌、升高丁酸产生菌的相对丰度,促进乳酸向丁酸转化,维持肠道微生态平衡稳定相关。
Objective:To investigate the influence of Zhengan Xifeng Decoction(镇肝熄风汤)on the intestinal flora in spontaneously hypertensive rats(SHRs).Methods:Fifty SHRs were randomly assigned into a model group,a benazepril(0.9 mg/kg)group,and low-,medium-,and high-dose Zhengan Xifeng Decoction(15,30,and 60 g/kg,respectively)groups.In addition,10 WKY rats were taken as the normal control group.The rats were administrated with corresponding drugs or distilled water for 8 weeks.After that,the blood pressure was measured,and the cecal and colonic feces were sampled for the sequencing of the 16 S rDNA V3-V4 amplicon.Results:Compared with the normal control group,the model group had elevated systolic blood pressure(SBP),diastolic blood pressure(DBP),and mean arterial blood pressure(MBP)(P<0.05),increased abundance of Turicibacter and decreased abundance of Clostridium in the cecum(P<0.05),and increased abundance of Lactobacillus and decreased abundance of Coprococcus,and Ruminococcus in the colon(P<0.05).Compared with the model group,benazepril and Zhengan Xifeng Decoction lowered the SBP,DBP,and MBP(P<0.05)and decreased the abundance of Proteobacteria.They decreased the relative abundance of Turicibacter capable of producing lactic acid and increased that of Clostridium capable of producing butyric acid in the cecum(P<0.05).Furthremore,they decreased the relative abundance of lactic acid-producing bacteria Lactobacillus and increased that of the butyric acid-producing bacteria Coprococuus and Ruminococcus in the colon(P<0.05).The alpha diversity increased in all the intervention groups.Beta diversity showed that the distribution of the flora in the intervention groups was more concentrated than that in the model group.Conclusion:Zhengan Xifeng Decoction may reduce the relative abundance of lactic acid-producing bacteria and increase the relative abundance of butyric acid-producing bacteria in rat intestine so as to promote the conversion of lactic acid to butyric acid,thereby maintaining the balance of intestinal microecology and lowering the blood pressure of SHRs.
