摘要:目的基于动物脑电技术研究四逆散干预对创伤后应激障碍(PTSD)所致大鼠睡眠障碍的影响。方法用随机数表法将SD大鼠分为5组,每组10只:对照组、模型组、阴性对照组、阳性对照组和实验组。除对照组外,其余4组均复制模型。对照组只埋置电极,正常饲养;模型组不接受治疗;阴性对照组灌胃等体积0.9%Na Cl;阳性对照组灌胃盐酸帕罗西汀溶液0.42 mg·m L^(-1);实验组灌胃四逆散水煎液(含生药0.24 g·m L^(-1));于应激造模前1 h灌胃给药10m L·kg^(-1)。每天灌服1次,共计7 d。第7天干预结束后18:00插上电缆,设定软件记录程序,次日记录08:00-20:00睡眠脑电,同时开启监控摄像系统。结果对照组大鼠脑电图波形整齐,均匀规律。模型组与对照组比较,大鼠脑电波波形模糊、频率高低夹杂。阴性对照组与模型组比较,变化特征与之相似。阳性对照组和实验组与阴性对照组比较,大鼠脑电波的波形较清晰,混杂的高频波明显减少,清晰易辩。结论四逆散对PTSD大鼠的睡眠时相有明显的正向调节作用。
Objective To study the effects of Sinisan intervention on rats with post traumatic stressdisorder( PTSD) and sleep disorder.Methods Fifty SD rats were divided equally into 5 groups: control group,model group,negative control group,positive control group and experimental group. All the groups of rats were implanted with electroencephalogram( EEG) electrodes,and recovered for 5 d,and inserted into the cable to adapt to 24 h at 8: 00 on the morning of the sixth day. The next day began measuring 1 d baseline EEG at 08: 00. Copied the date of the end of the EEG detection model. The control group did not copy the model,buried electrode,normal feeding. Model group,repetitive PTSDmodel was induced by current stimulation,but not treated. Negative control group,equal volume of 0. 9% Na Cl. Positive control group was given paroxetine hydrochloride 0. 42 mg · m L^(-1) and experimental groupwas given Sinisan( Containing crude drug 0. 24 g·m L^(-1)). It given to drugs one hour before the model establishment. The rats were administered with10 m L·kg^(-1),once a day,for a total of 7 d. The cable were plugged at18: 00 on the seventh day,set the software recording program. The next day recorded sleep EEG from 08: 00 to 20: 00,while opening the surveillancecamera system. Results Compared with the control group,the brain wave of model group was fuzzy and the frequency was high or low. Compared with model group,similar variation characteristics of the negative control group. Compared with negative control group,waveform was more clear in the positive control group and experimental group,the hybrid high frequency wave significantly reduced,clear and easy to debate. The waveform characteristics of rats in the experimental group were similar to those in the positive control group. Conclusion The Sinisan has significant positive effect on the sleep phase of rats with post traumatic stress.
