摘要:研究党参水煎液对大鼠实验性溃疡性结肠炎(UC)的保护作用并探讨其抗炎作用机制。将40只大鼠随机分为正常组、模型组、阳性对照组(柳氮磺胺嘧啶,SASP;0.3 g/kg)和党参水煎液组(18 g/kg),用三硝基苯磺酸(TNBS)/乙醇联合灌肠建立大鼠UC模型,连续给药21 d,观察大鼠一般情况,进行疾病活动指数(DAI)和结肠黏膜损伤指数(CMDI)评分;H.E染色观察结肠组织形态;生化法检测结肠中超氧化物歧化酶(SOD)和丙二醛(MDA)含量;RT-PCR法检测结肠组织白介素-6(IL-6)、白介素-10(IL-10)及肿瘤坏死因子(TNF-α)的mRNA表达;免疫组化法检测结肠组织中NF-κB的蛋白表达。结果表明,党参水煎液能显著降低UC大鼠DAI评分、CMDI评分(P<0.01);改善结肠黏膜形态;升高结肠组织中SOD活力,降低MDA含量(P<0.01);抑制结肠组织中IL-6、TNF-αmRNA水平,促进IL-10 mRNA表达(P<0.01);同时降低结肠中NF-κB的蛋白表达(P<0.01)。提示党参水煎液对大鼠溃疡性结肠炎具有保护作用,其抗炎机制可能与抗脂质过氧化,抑制NF-κB的表达而减少促炎因子的释放有关。
To study the protective effect of Codonopsis pilosula decoction on ulcerative colitis(UC)in rats and to explore its anti-inflammatory mechanism.Forty rats were randomly divided into normal group,model group,positive control group(sulphasalazine,SASP;0.3 g·kg^-1)and Codonopsis decoction group(18 g·kg^-1).Trinitrobenzene sulfonic acid(TNBS)/ethanol combined with enema was used to establish a rat UC model.After 21 days of continuous administration,the general condition of the rats was observed and the disease activity index(DAI)and colonic mucosal injury index(CMDI)scores were obtained.Colon tissue morphology was observed by staining.The contents of superoxide dismutase(SOD)and malondialdehyde(MDA)in the colon were detected by biochemical methods.The contents of inflammatory cytokines interleukin-6(IL-6),interleukin-10(IL-10)and tumor necrosis factor(TNF-α)in colon tissues were determined by RT-PCR.The protein expression of NF-κB in colon tissues was determined by immunohistochemistry.DAI,CMDI and colonic mucosa morphology of UC rats were significantly decreased and improved in Codonopsis decoction(P<0.01).Furthermore,the SOD activity in colon tissues was increased and MDA content was decreased(P<0.01).Codonopsis decoction significantly reduced IL-6 and TNF-αm RNA levels in colon tissues and increased IL-10 mRNA levels(P<0.01).Meanwhile,the protein expression of NF-κB in colon was significantly decreased(P<0.01).Codonopsis pilosula decoction has protective effects on ulcerative colitis in rats,and its anti-inflammatory mechanism may be related to anti-lipid peroxidation,inhibition of NF-κB expression then reducing the release of pro-inflammatory factors.
