摘要:目的:探讨环氧合酶m RNA(cox-2 m RNA)表达在合募配穴针刺防治急性胃炎中的效应机制。方法:将60只SD大鼠(180-220 g,雄性)按计算机随机生成的随机数字表法分为空白对照组、抓取组、模型组、中脘穴组、后三里穴组、合募配穴组(中脘穴+后三里穴组)6组,每组10只。采用烈性白酒灌胃法复制急性胃炎大鼠模型。针刺组大鼠依照分组要求取相应穴位,均采用捻转平补平泻法针刺(捻转幅度180°,频率60-80次/min),1次/d,每次20 min,造模前连续针刺6 d,造模后,连续针刺3 d。治疗结束后用实时荧光定量聚合酶链式反应(q RT-PCR)技术检测胃和下丘脑组织cox-2 m RNA的表达变化。结果:与空白对照组相比,抓取组胃组织与下丘脑组织中cox-2m RNA表达水平显著升高(P<0.05);与抓取组相比,模型组胃组织与下丘脑组织中cox-2 m RNA表达水平有极显著性升高(P<0.01);与模型组相比,3个针刺组胃组织与下丘脑组织中cox-2 m RNA的表达均有极显著性下调(P<0.01);3个针刺组组间比较,胃组织中cox-2 m RNA的表达无显著性差异(P>0.05),合募配穴组下丘脑组织cox-2m RNA的表达量最低,与中脘穴组相比有极显著性差异(P<0.01),与后三里穴组相比有显著性差异(P<0.05)。结论:针刺防治酒精性胃炎的机制可能与通过抑制胃、下丘脑组织cox-2 m RNA表达密切相关;在降低下丘脑cox-2m RNA表达水平方面,配穴(合募配穴)效果优于单穴(后三里穴或中脘穴)作用,具有一定特异性。
Objective: To study the mechanism of hypothalamic and gastric tissue cox-2 mRNA expression in treatment of acute gastritis of rats by acupuncture at Ho Points and Front-Mu Points. Methods: 60 SD rats(180-220 g, male) were randomly divided into six groups applying the method of random number table generated by computer as follows, blank control group, grab group,model group, Zhongwan Points(RN 12) group, Housanli Points(ST 36) group and Ho-Mu combination acupoints group(Zhongwan matching Housanli), with 10 rats in each group. Adopt potent liquor to duplicate rat model of acute gastritis. Each treatment group in accordance with the relevant requirements to take corresponding acupoints, and adopt acupuncture therapy of twirling and mild reinforcing-reducing method(twist amplitude of 180°, the frequency of 60-80 times/min) once daily at 20 minutes every time.Acupuncture last continuously 6 days and 3 days respectively, before and after duplicating rat model. Then cox-2 mRNA expression in gastric tissue and hypothalamus were detected with real-time fluorescence quantitative polymerase chain reaction(q RT-PCR) technology. Results: The expression of cox-2 mRNA in gastric tissue and hypothalamus all increased significantly(P〈0.05) in grab group compared with blank control group; The expression of cox-2 mRNA in gastric tissue and hypothalamus all increased marked significantly(P〈0.01) in model group compared with grab group; The expression of cox-2 mRNA in gastric tissue and hypothalamus all decreased marked significantly(P〈0.01) in three acupuncture groups compared with model group; Within three acupuncture groups, there were no significant difference(P〉0.05) in the expression of cox-2 mRNA in gastric tissue, the expression of cox-2 mRNA in hypothalamus was lowest compared with Housanli Points group and Zhongwan Points group(P〈0.01 or P〈0.05). Conclusion:The mechanism of acupuncture on the treatment and prevention of alcoholic gastritis may be related with the inhibition of cox-2 mRNA expression of gastric tissue and hypothalamus; In reducing the cox-2 mRNA expression on hypothalamus, the effect of acupoints with specificity(Ho-Mu combination acupoint) was better than the single point(Zhongwan and Housanli).
