摘要:目的观察淫羊藿苷对激素抵抗型肾病综合征大鼠的疗效及可能机制的研究。方法健康雄性SD大鼠60只,空白组10只,50只制备成功的SRNS模型大鼠随机分为模型组、激素组、淫羊藿苷组、联合组各10只,连续给药6周。给药2周、4周和6周测定各组大鼠体重、24 h尿蛋白含量;末次给药结束后,检测相关生化指标;采用HE、Masson、PAS染色观察大鼠肾组织病理形态学变化情况;Tunel染色观察大鼠肾组织细胞凋亡情况;免疫荧光法检测p-ERK1/2表达情况;通过Western blot检测SRNS大鼠的Ras、A-Raf、B-Raf、ERK1/2、p-ERK1/2的蛋白含量。结果与空白组相比,模型组24 h尿蛋白量升高,ALB、Scr水平降低,TC、TG、BUN水平升高,肾小球硬化评分升高,肾组织细胞凋亡率升高,Ras、A-Raf、B-Raf、ERK1/2、p-ERK1/2蛋白表达增加(P<0.05);与模型组相比,泼尼松组和淫羊藿苷组大鼠上述异常无明显改善,联合组24 h尿蛋白量降低,ALB、Scr水平升高,TC、TG、BUN水平降低,肾组织损伤、肾组织细胞凋亡率降低,Ras、A-Raf、B-Raf、ERK1/2、p-ERK1/2蛋白表达降低,差异有统计学意义(P<0.05)。结论淫羊藿苷可缓解SRNS大鼠肾组织损伤,保护肾功能,这可能与淫羊藿苷抑制Ras/Raf/ERK 1/2信号通路,降低肾组织细胞凋亡有关。
Objective To observe the effect of Icariin on hormone resistant nephrotic syndrome rats and its possible mechanism.Methods 60 healthy male SD rats,10 in the blank group and 50 in the successfully prepared SRNS model rats were randomly di?vided into model group,hormone group,icariin group and combined group,with 10 rats in each group,and were administered continuously for 6 weeks.The body weight and 24 h urinary protein content of rats were measured at 2,4 and 6 weeks after ad?ministration;After the last administration,relevant biochemical indexes were detected;He,Masson and PAS staining were used to observe the pathomorphological changes of rat kidney tissue;TUNEL staining was used to observe the apoptosis of renal cells in rats;The expression of p-ERK1/2 was detected by immunofluorescence;The protein contents of RAS,A-Raf,B-Raf,ERK1/2 and p-ERK1/2 in SRNS rats were detected by Western blot.Results compared with the blank group,the 24-h uri?nary protein content of the model group increased,the ALB and Scr level decreased,the TC,TG,BUN levels increased,the glo?merular sclerosis score increased,the apoptosis rate of renal tissue increased,and the expression of RAS,A-Raf,B-Raf,erk1/2,p-ERK1/2 protein increased(P<0.05).Compared with the model group,the above abnormalities of rats in the pred?nisone group and icariin group were not significantly improved.In the combined group,24-h urinary protein decreased,ALB and Scr levels increased,TC,TG,BUN levels decreased,renal tissue injury and renal tissue apoptosis rate decreased,and RAS,A-Raf,B-Raf,erk1/2,p-ERK1/2 protein expression decreased,the difference was statistically significant(P<0.05).Conclusion icariin can alleviate renal tissue injury and protect renal function in SRNS rats,which may be related to icariin inhib?iting Ras/raf/erk 1/2 signal pathway and reducing renal cell apoptosis.
