摘要:目的观察老年性痴呆(AD)大鼠学习记忆及皮层ChAT阳性细胞的表达和病理形态的变化,探讨寿聪胶囊的防治机理。方法选用Wistar雄性大鼠随机分成五组:空白对照组、模型对照组、脑复康组及寿聪胶囊低、高剂量组。用D-半乳糖和东莨菪碱制备复合AD大鼠模型,应用中药复方寿聪胶囊进行防治。用跳台法测试大鼠的学习记忆能力,免疫组化法观察大脑皮层区ChAT阳性细胞的表达及皮层病理形态学改变。结果寿聪胶囊组跳台潜伏时间明显延长,触电错误次数显著减少,大脑皮层ChAT的阳性细胞数目显著增多,脑组织病理损害明显减轻,与模型组相比较P<0.01,与脑复康组相似(P>0.05)。结论寿聪胶囊能改善AD大鼠的学习记忆并减轻皮层组织的损害,可能与寿聪胶囊保护胆碱能神经系统的功能,提高ChAT的活性,促进Ach的合成有关。
Objective To study the effect of ShouCong capsule on learning and memory ability,ChAT and pathological changes of cerebra cortex in rats with Alzheimer's disease.Methods Male Wistar rats were randomly divided into 5 groups: blank control group,model group,NaoFuKang group,ShouCong capsule(low,high dose)group.Alzheimer's disease(AD) model rats were made by injection with D-galactose on the back of the neck,then injection with scopolamin in the intraperiton.Learning and memory ability of rats was evaluated with a series of tasks to find a hidden plation in dark box,the tissue pathological changes under light microscope and choline acetyltransferase(ChAT) by electron lens using immunohistochemistry method were observed.Results In low and high dose ShouCong capsule groups,the electric latent period was lengthened while 5-minute error frequency was decreased;the pathological injury of dementia rats was respectively lessened.The number of ChAT positive neurons in cerebral cortex was significantly increased compared with that in the model group(P0.01),but there was not cerebral significantly increased compared with NaoFuKang group(P0.05).Conclusion ShouCong capsule can greatly improve learning and memory capacity,lessen the pathological injury of dementia rats,and the mechanism is possibly relevant with the protection of cholinergic system,enhancment of ChAT evel to further synthesize Ach.
