摘要:目的探讨消定膏通过核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)/核因子κB(NF-κB)/半胱天冬酶1(Caspase-1)通路抑制细胞焦亡,对急性闭合性软组织损伤的治疗效果。方法选取60只SPF级SD大鼠,随机分为4组:空白组、模型组、阳性对照组(使用奇正消痛贴膏)和消定膏组(外用敷贴)。空白组不做任何处理,其余组在制备急性闭合性软组织损伤模型后,进行相应干预,治疗周期为7 d。通过损伤症状评分评估肿胀、疼痛、活动受限等情况。苏木精-伊红(HE)染色观察大鼠腿部肌肉组织组织病理学变化。ELISA检测血清炎症因子IL-1β、TNF-α水平。TUNEL染色观察细胞焦亡情况。Western blot和免疫组化分析NLRP3、NF-κB、p65、Caspase-1蛋白表达水平,评估消定膏的疗效及作用机制。结果与模型组相比,消定膏组大鼠的肿胀程度、外观变化、活动情况及疼痛行为明显改善(均P<0.05),组织病理学结果显示炎症细胞浸润减少,肌纤维排列趋于正常;血清炎症因子IL-1β、TNF-α水平显著降低(均P<0.05);焦亡阳性细胞数及焦亡阳性率明显下降(均P<0.05);NLRP3、NF-κB、p65及Caspase-1蛋白表达明显降低(均P<0.05)。结论消定膏通过抑制NLRP3炎症小体活化及NF-κB信号通路,阻断Caspase-1依赖性细胞焦亡进程,减少IL-1β、TNF-α炎症因子释放,从而减轻急性软组织损伤后的炎症反应,促进组织修复。
Objective To investigate the molecular mechanism of Xiaoding Ointment in inhibiting pyroptosis through nucleotide binding oligomerization of domain like receptor protein 3(NLRP3)/nuclear factor kappa B(NF-κB)/caspase-1 pathway,and to analyze its therapeutic effect on acute closed soft tissue injury.Methods Sixty SPF SD rats were selected and randomly divided into four groups:blank group,model group,positive control group(Qizheng Xiaotong plaster),and Xiaoding Ointment group(topical application).The blank group did not receive any treatment,while the other groups received corresponding intervention treatment after preparing the acute closed soft tissue injury model,with a treatment period of seven days.The conditions such as swelling,pain and limited movement were evaluated through the injury symptom score.Hematoxylin-eosin(HE)staining was used to observe the histopathological changes of rat leg muscle tissue.ELISA was used to detect the levels of serum inflammatory factors IL-1β and TNF-α.TUNEL staining was used to observe the pyroptosis of cells.The expression levels of NLRP3,NF-κB,p65 and Caspase-1 proteins were analyzed by Western blotting and immunohistochemistry to evaluate the therapeutic effect and mechanism of action of Xiaoding Ointment.Results Compared with the model group,the swelling degree,appearance changes,activity and pain behavior of rats in the Xiaoding Ointment group were significantly improved(all P
