摘要:目的:建立一种稳定的、可操作性强的病证结合大鼠帕金森病(PD)模型,以满足研究药物对PD治疗评价的需求。方法:采用6-羟基多巴胺脑部立体注射法构建PD动物模型,同时设立正常对照组、假手术组、模型组、药物(复方地黄方)组,从病因、症状、客观指标、药物反证等方面对PD大鼠模型进行综合判断,验证该模型的中医证候属性。结果:模型组大鼠的神经行为学、氧化应激、TH m RNA及蛋白、阳性细胞等指标与正常对照组比较,差异均有统计学意义(P<0.01)。药物组可明显改善大鼠的主观症状和上述客观指标,差异有统计学意义(P<0.01)。结论:6-羟基多巴胺大鼠PD模型的中医证候属性是风阳内动证。
Objective: To establish a steady and simplify combination of disease and syndrome rat model with Parkinson's disease(PD), and meet the demand of drug study on treatment and evaluation of PD. Methods: The animal PD model was established by brain stereotaxic injection with 6-OHDA. The rats were divided into normal control group, sham operation group, model group and drug group. The PD model and its syndrome property were validated by a synthetic judgment from the aspects of etiology, symptoms, objective index, drugs counterevidence and so on. Results: The nerve etiology, oxidative stress, TH m RNA and protein and positive cell in model group were significantly different compared with the normal control group(P〈0.01). Drug group could significantly improve the subjective symptoms and objective indexes of rats mentioned above, the difference was statistically significant(P〈0.01). Conclusion: The TCM syndrome of PD 6-OHDA model rats was syndrome of internal stirring of wind yang.
