摘要:目的:探讨归芪益元膏对重离子辐射旁效应损伤大鼠Bax、Bcl-2及外周血象的影响。方法:Wistar雄性大鼠随机分为3组,正常对照组、辐射组和中药组。中药组预先给予归芪益元膏灌胃2周(3.28g·kg-1·d-1)后辐射组和中药组大鼠右肺予2Gy12C6+离子束单次照射,正常对照组不予照射。照射后24h处死各组大鼠,Western Blot及Q-PCR检测大鼠肺肾组织中Bax、Bcl-2蛋白及m RNA表达,检测外周血象。结果:与正常对照组比较,辐射组大鼠肺肾组织中Bax蛋白及m RNA表达上调(P<0.05),Bcl-2蛋白及m RNA表达下调(P<0.05),白细胞、红细胞、血小板、血红蛋白含量均下降(P<0.05)。与辐射组比较,中药组大鼠肺、肾组织中Bax蛋白及m RNA表达下调(P<0.05),Bcl-2蛋白及m RNA表达上调(P<0.05),白细胞、红细胞、血小板、血红蛋白含量均升高(P<0.05)。结论:归芪益元膏可以减轻重离子辐射旁效应诱导的肺、肾细胞凋亡,保护外周血象,从而发挥其对重离子辐射旁效应损伤的防护作用。
Objective: To explore the effects of Guiqi Yiyuan Ointment on Bax, Bcl-2 and peripheral blood of rats damaged by heavy ion radiation induced bystander effects. Methods: Male Wistar rats were randomly divided into three groups, the normal control group, radiation group and the Chinese materia medica group. Rats in the Chinese materia medica group were given Guiqi Yiyuan Ointment by gavage for two weeks (3.28g·kg^-1·d^-1) in advance. After two weeks, the right lurig of the rats of the radiation group and the Chinese materia medica group was radiated once by 2Gy^12C6^+ ion. The normal control group was not radiated. After radiation for 24h, all groups of rats were executed. The amount of protein and mRNA expression of Bax and Bcl-2 in lung and kidney was examined with Western Blot and Q-PCR. Peripheral hemogram were measured. Results: Compared with the normal control group, the amount of Bax protein and mRNA expression of the radiation group increased significantly (P〈0.05). The amount of Bcl-2 protein and mRNA expression of the radiation group decreased significantly (P〈0.05). The contents of leukocyte, erythrocyte, platelet and hemoglobin in peripheral blood decreased significantly (P〈0.05). Compared with the radiation group, the amount of Bax protein and mRNA expression of the Chinese materia medica group decreased significantly (P〈0.05). While the amount of Bcl-2 protein and mRNA expression of the Chinese materia medica group increased significantly (P〈0.05). The contents of leukocyte, erythrocyte, platelet and hemoglobin in peripheral blood increased significantly (P〈0.05). Conclusion: Guiqi Yiyuan Ointment could reduce the apoptosis of lung and kidney cells induced by heavy ion radiation bystander effects and protect peripheral blood cells, so as to exert its protective effect on the bystander effects of heavy ion radiation.
