摘要:目的:观察当归挥发油对吗啡依赖小鼠戒断症状及单胺类神经递质的影响。方法:90只昆明小鼠随机均分为空白组、模型组、可乐定组(0.6 mg·kg-1)和当归挥发油低、中、高剂量组(20,40,80 mg·kg-1)。采用剂量递增法建立小鼠吗啡依赖模型,观察纳洛酮催促戒断后30 min内的小鼠的戒断症状(跳跃数、扭体次数、抓脸次数和体重减少值);采用酶联免疫吸附法测量下丘脑和血浆中NE、DA和5-HT的含量。结果:当归挥发油呈剂量依赖地降低吗啡依赖小鼠戒断后跳跃次数(F=22.1,P<0.01)、扭体次数(F=15.1,P<0.01)、抓脸次数(F=16.4,P<0.01)及体质量减少百分值(F=11.1,P<0.01),同时显著性降低下丘脑和血浆中NE、DA和5-HT水平[(F=14.7,P<0.01;F=16.3,P<0.001;F=11.7,P<0.01);(F=10.4,P<0.001;F=13.5,P<0.001;F=9.4,P<0.01)]。结论:当归挥发油能够有效缓解小鼠吗啡戒断症状,其机制可能与抑制下丘脑和血浆中单胺类神经递质的过度释放有关。
OBJECTIVE To investigate the effects of Angelica naphtha against withdrawal symptoms and monoamine neurotransmitter in morphine-dependent mice.METHODS A total of 90 kunming mice were randomly divided into control group,model group,clonidine group(0.6 mg·kg-1)and Angelica naphtha groups(20,40,80 mg·kg-1).Morphine dependence model was established by gradually increasing morphine dose in mice.Morphine withdrawal syndrome was precipitated by naloxone and withdrawal symptoms(jump,writhing,scratching and weight loss)were observed and evaluated for 30 min.The level of norepinephrine(NE),dopamine(DA)and serotonin(5-HT)of mice were determined with ELISA assay.RESULTS The jump,writhing,scratching and weight loss of morphine withdrawn mice were reduced by Angelica naphtha in a dose dependent manner(F=22.1,P〈0.01;F=15.1,P〈0.01;F=16.4,P〈0.01),levels of NE,DA and 5-HT in hypothalamus and plasma were also significantly decreased[(F=14.7,P〈0.01;F=16.3,P〈0.001;F=11.7,P〈0.01);(F=10.4,P〈0.001;F=13.5,P〈0.001;F=9.4,P〈0.01)].CONCLUSION Angelica naphtha can effectively alleviate symptoms of morphine withdrawal in mice,which may be related with inhibition of excessive release of monoamine neurotransmitters in hypothalamus and plasma.
