摘要:目的探讨脾气虚大鼠骨骼肌组织钙调蛋白信号通路中钙调蛋白(Ca M)、钙调素依赖蛋白激酶(Ca MK)Ⅱ基因表达变化规律。方法受试动物随机分为空白组、脾气虚模型组(7 d、14 d、21 d组),每组10只。除空白组外,其余受试动物采用复合法(苦寒破气法、力竭法及饥饮失常法)成功建立脾气虚证大鼠模型后,在观察各组大鼠一般生存状态、脾虚证宏观证候积分、平均每日摄食量、平均每日体重增加量和负重游泳耐力的同时,采用实时荧光定量PCR技术检测骨骼肌组织Ca M信号通路中Ca M、Ca MKⅡ基因表达水平的变化。结果与空白组比较,脾气虚7 d、14 d、21 d组大鼠一般生存状况较差,脾虚宏观证候积分显著升高(P<0.05),平均每日摄食量、平均每日体重增加量和负重游泳耐力降低(P<0.05),骨骼肌组织Ca M、Ca MKⅡ基因相对表达量显著降低,且以脾气虚模型21 d组变化显著(P<0.05)。结论脾气虚大鼠骨骼肌组织Ca M信号通路中Ca M、Ca MKⅡ基因表现为低表达水平。
Objective To research the expressions of Ca M,Ca MKⅡ mRNA in calmodulin signaling pathways in skeletal muscle of rats with spleen-qi deficiency. Methods Rats were randomly divided into normal,spleen-qi deficient model groups( observed on 7 d,14 d and 21 d),10 rats in each. The spleen-qi deficient model rats were made by rhubarb,exhaustive and hungry method; then the general condition,macroscopic spleen deficiency syndrome integral,the average food intake,daily weight gain and loading swimming endurance were evaluated,and the expressions of Ca M,Ca MKⅡ mRNA in skeletal muscle tissue were evaluated by real time fluorescent quantitative PCR. Results Compared with those of normal group,the general of survival was poor,macroscopic spleen deficiency syndrome integral was increased( P <0. 05),the average food intake,average daily weight gain and loading swimming endurance were decreased( P < 0. 05),and relative expression of Ca M,Ca MKⅡ mRNA in skeletal muscle tissue were decreased in model group,this difference was obvious on spleen-qi deficient model 21 d group( P < 0. 05). Conclusions The expressions of Ca M,Ca MKⅡ mRNA are decreased in calmodulin signaling pathways in skeletal muscle of rats with spleen-qi deficiency.
