摘要:骨质疏松症是一种由多种原因引发的代谢性骨病,主要特征是以骨密度和微结构受到损害,导致骨的脆性增加,骨折发生风险上升。其发病机制错综复杂,多种因素相互作用、相互影响,共同推动了疾病进展。泛素化调节是一种关键的蛋白质修饰机制,在骨代谢进程里有着不可或缺的地位。目前研究表明,泛素特异性蛋白酶可通过调节成骨细胞、破骨细胞的分化及功能,进而影响骨重建过程。而泛素化作为一种新型的蛋白质修饰方式,为治疗骨质疏松症提供了更多可能。本文综述了泛素化修饰的一系列基本过程、相关酶类以及其在骨质疏松发病机制中的作用(对成骨和破骨细胞功能的调节)、与信号通路的相互作用以及在骨重建过程中的影响。更加深入地认识泛素化调节理论对于骨质疏松的发病机制有着重要作用,将为开发新的治疗策略提供更好的理论依据。
Osteoporosis is a metabolic bone disease caused by multiple factors.Its main characteristics are the impairment of bone mineral density and microstructure,which leads to an increase in bone fragility and the risk of fractures.The pathogenesis of this disease is extremely complex,with multiple factors interacting and influencing each other,jointly promoting the progression of the disease.Ubiquitination regulation is a crucial protein modification mechanism and plays an indispensable role in the process of bone metabolism.Current research indicates that ubiquitin-specific proteases affects the bone remodeling process by regulating the differentiation and function of osteoblasts and osteoclasts.Ubiquitination,as a novel protein modification method,offers more possibilities for the treatment of osteoporosis.This paper reviews a series of basic processes of ubiquitination modification,related enzymes,and the role of ubiquitination in the pathogenesis of osteoporosis,including the regulation of the functions of osteoblasts and osteoclasts,the interaction with signal pathways,and the impact on the bone remodeling process.A more in-depth understanding of the important role of ubiquitination regulation theory in the pathogenesis of osteoporosis provides a better theoretical basis for the development of new treatment strategies.
