摘要:目的:运用大数据和网络药理学方法分析清肺排毒汤对三种冠状病毒感染的SARS、MERS和COVID-19的共性作用机制,探讨清肺排毒汤在冠状病毒感染治疗方面的潜在应用价值。方法:通过TCMSP、TCMID和GeneCards数据库收集清肺排毒汤组方药物化学成分作用靶点和SARS、MERS和COVID-19疾病相关靶点,应用Cytoscape软件构建清肺排毒汤治疗SARS、MERS和COVID-19的化学成分-靶点-疾病交互网络,通过STRING数据库构建蛋白互作网络,运用DAVID数据库进行GO和KEGG分析,探讨清肺排毒汤治疗SARS、MERS和COVID-19的潜在共性作用。结果:SARS疾病相关靶点7 820个、MERS疾病相关靶点1 853个、COVID-19疾病相关靶点259个,其中与三种疾病相关的共性靶点148个;从清肺排毒汤中筛选出化学成分246个,对应靶点337个,其中44个靶点和191个化学成分与上述三种疾病相关,其中TP53、CASP3、IL-6、MAPK8、MAPK1、IL-10、CCL2、IL-1β、CXCL8、MAPK14等可能是清肺排毒汤治疗上述三种疾病的潜在共性靶点,槲皮素、木犀草素、汉黄芩素、山柰酚、柚皮素等可能是清肺排毒汤治疗上述三种疾病的潜在共性成分,这些化学成分多为黄酮类。影响血管生成、免疫反应、一氧化氮合成及细胞增殖与凋亡等生物过程可能是清肺排毒汤对三种疾病的潜在共性调节机制。结论:清肺排毒汤对SARS、MERS和COVID-19三种疾病具有潜在共性作用机制,提示该药不仅对COVID-19具有较好的疗效,可能在治疗SARS和MERS方面也具有一定的应用价值。
Objective: Big data and network pharmacology were used to analyze the common action mechanism of Qingfeipaidu decoction in the treatment of SARS, MERS and COVID-19, and to explore the potential application value of Qingfeipaidu decoction in the treatment of coronavirus infection. Methods: The TCMSP, TCMID GeneCards database were used to collect the chemical composition action target and disease-related targets for SARS, MERS and COVID-19. The Cytoscape software was used to construct the chemical component-target-disease interaction network of Qingfeipaidu decoction for the treatment of SARS, MERS and COVID-19. STRING database was used to build the protein interaction network, GO and KEGG analysis was constructed by DAVID database, to explore the potential common action of Qingfeipaidu decoction in the treatment of SARS, MERS and COVID-19. Results: There were 7820 disease-related targets for SARS, 1853 disease-related targets for MERS and 259 disease-related targets for covid-19, while 148 common targets for three diseases were screened. There were 246 chemical components and corresponding 337 targets in Qingfeipaidu decoction, among which 44 targets and 191 chemical components were related to SARS, MERS and COVID-19. TP53, CASP3, IL6, MAPK8, MAPK1, IL10, CCL2, IL1 B, CXCL8, MAPK14, etc., might be the potential common targets of Qingfeipaidu decoction in the treatment of these three diseases. Quercetin, luteolin, baicalin, kaempferol, naringenin might be the main potential common chemical components of Qingfeipaidu decoction in the treatment of these three diseases, and these compounds were basically flavonoids. Biological angiogenesis, immune response, nitric oxide synthesis, cell proliferation and cell apoptosis might be the potential common regulatory mechanisms of Qingfeipaidu decoction in the treatment of SARS, MERS and COVID-19. Conclusion: It is possible that Qingfeipaidu decoction may have potential common action mechanism in the treatment of SARS, MERS and COVID-19. The results suggest that it has a good effect on COVID-19, and good application value in the treatment of SARS and MERS.
