摘要:目的:探讨当归红芪超滤物对急性心肌梗死大鼠超氧化物歧化酶(SOD)活性、丙二醛(MDA)、乳酸脱氢酶同工酶(LDH1)含量和热休克蛋白(Hsp70)表达的影响,阐明当归红芪超滤物对梗死心肌的保护作用。方法:结扎大鼠左前降支建立心梗模型。将60只雄性Wstiar大鼠随机分成5组,每组12只,分别为造模组(MI组)、假手术组(SH组)、低剂量组(LD组)、中剂量组(MD组)、大剂量组(HD组),各组分别于治疗后(24h、7d)断头处死,分别测外周血SOD、MDA、LDH1和采用RT-PCR技术检测大鼠心肌组织HSP70 mRNA的表达。结果:与MI相比,LD组、MD组、HD组均降低了ST段抬高、MDA、LDH1水平(P<0.05,P<0.01),增强了SOD活性、HSP70的表达(P<0.05,P<0.01)。结论:当归红芪超滤物对急性心肌缺血大鼠具有明显的保护作用。
Objective: To investigate the effect of the ultra-filtration extract mixture from Angelica and Hedysari,radix on SOD,MDA,LDH1 and HSP70 expression in acute myocardial infarction(AMI) rat,and clarify its protection on cardiomyocytes.Methods: AMI were established with the ligation of left anterior descending artery.80 male Wister rats were randomly divided as model(MI) group,sham-operation(SH) group,low dosage(LD) group,middle dosage(MD) and high dosage(HD) group,each group included 16 animals.The rats were sacrificed at 24 hours,7 days after treatment,respectively.The expression of peripheral blood serum SOD,MDA,LDH1 and the expressions of Hsp70 mRNA in cardiomyocytes were measured by RT-PCR.Results: Compared with myocardial infarction group,ST segment elevations,MDA,LDH1 were significantly decreased(P〈0.05,P〈0.01) and active SOD,the expressions of Hsp70 mRNA in cardiomyocytes were increased(P〈0.05,P〈0.01).Conclusion: The ultra-filtration extract mixture can protect myocardium on AMI.
