摘要:目的:探讨黄芪多糖对大鼠脑缺血再灌注损伤后脑组织Bcl-2、Bax蛋白及NO含量的影响。方法:将SPF级Wistar大鼠72只分为假手术组、模型组、阳性对照组及黄芪多糖高、中、低剂量组,采用改良的线栓法制作大鼠脑缺血再灌注模型,各药物组分别给予相应药物15天后采用免疫组化法检测Bax、Bcl-2蛋白含量的表达,采用硝酸盐还原酶法检测NO蛋白含量的表达。结果:1)Bcl-2、Bax的含量模型组较假手术组表达明显增加;与模型组比较,Bcl-2表达黄芪多糖高、中剂量组增加(P<0.05);Bax含量黄芪多糖高剂量组表达显著减少(P<0.05)。与假手术组比较,模型组Bcl-2/Bax比值明显降低,阳性对照组与黄芪多糖各剂量组比较比值升高。2)与假手术组比较,模型组大鼠脑组织中NO的水平明显升高(P<0.05),与模型组比较,黄芪多糖高剂量组中NO含量明显降低(P<0.05)。结论:黄芪多糖可增加凋亡抑制基因Bcl-2蛋白和降低凋亡促进基因Bax蛋白的表达,通过增加Bcl-2/Bax比例,降低NO含量,减少细胞凋亡的凋亡程度。
Objective: To explore the influence of astragalus polysaccharides on the contents of Bcl-2, Bax protein and NO in the brain tissue of the rats after cerebral ischemia reperfusion. Methods: Seventy-two SPF rats were randomized into sham operation group, the model group, positive control group, high, moderate and low doses groups of astragalus polysaccharides, the rat model was prepared by reformed middle cerebral artery occlusion, the expressions of Bax protein and Bcl-2 protein were detected with immunohistochemical method after different medication groups were administered with the corresponding herbs respectively, the expressions of NO protein was detected by nitrate reductase method. Results: 1) The contents of Bcl-2 and Bax in the model groups increased notably compared with sham operation group; compared with the model group, the expressions of Bcl-2 increased in high and moderate doses groups of astragalus polysaccharides(P<0.05); the contents of Bax protein decreased significantly in high dose group of astragalus polysaccharides(P<0.05). Compared with sham operation group, the ratio of Bcl-2/Bax lowered notably in the model group, it rose when positive control group was compared with different doses groups of astragalus polysaccharides. 2) Compared with sham operation group, the level of NO raised remarkably in the model group(P<0.05), compared with the model group, the contents of NO decreased evidently in the high dose group of astragalus polysaccharides(P<0.05). Conclusion: Astragalus polysaccharides could improve the expressions of Bcl-2 and Bax protein; it could reduce the degrees of cellular apoptosis by increasing the ratio of Bcl-2/Bax, and decreasing the contents of NO.
