摘要：目的:对吴茱萸生物碱进行提取纯化,并测定其含量,为中药外用制剂提供天然透皮促进剂。方法:利用吴茱萸碱和吴茱萸次碱的溶解性,采用浓度为70%乙醇提取吴茱萸生物碱,用中性氧化铝柱层析进行纯化,采用高效液相色谱法测定其含量。结果:吴茱萸碱的回归方程:y=5501x+35.37(r=0.999 5),线性范围为0.027-1.350μg,线性关系良好;吴茱萸次碱回归方程:y=1970x-4.018(r=0.999 5),线性范围为0.021 4-1.070 0μg,线性关系良好。吴茱萸总生物碱含量可达到86.6%。结论:该提取纯化方法所得样品纯度高,达到了提取有效部位要求,可为中药外用制剂选择天然透皮促进剂提供参考。
Objective: Total alkaloids in Evodia rutaecarpa was extracted and purified, and determination was performed by HPLC to provide high purity natural penetration enhancers for external application of Chinese herbal medicine. Methods:Total alkaloids were extracted by 70% of ethanol, purified by neutral alumina column chromatography and determined by HPLC. Results: The calibration curve of the evodiamine and rutaecarpine showed good linearity in the range of 0.027-1.350μg and 0.021 4-1.070 0 μg, the regression equation was y=5501x+35.37(r=0.999 5) and y=1970x-4.018(r=0.999 5), and the average recovery was 101.1% with RSD of 2.20% and 100.5% with RSD of 1.63%, respectively. Total alkaloids were more than 80% after extraction and purification. Conclusion: The high purity of total alkaloids in Evodia rutaecarpa obtained with this way meet the need of the effective parts, and could provide a reference for choose natural penetration enhancers for external application of Chinese herbal medicine.
