摘要:目的:研究当归贝母苦参丸加味方对MFC胃癌荷瘤小鼠瘤组织VEGFA、MMP13和TGF-β表达的影响。方法:建立MFC胃癌荷瘤小鼠模型进行体内抗肿瘤试验,将筛选合格的48只MFC胃癌荷瘤小鼠随机分为模型组、顺铂阳性对照组、加味当归贝母苦参丸高、低剂量组、顺铂联合加味当归贝母苦参丸高、低剂量组,连续灌胃给药14 d,治疗结束后,处死大鼠,RT-q PCR和免疫组织化学技术分别检测肿瘤组织中VEGFA、MMP13和TGF-β的表达。结果:与模型组相比,各用药组中VEGFA、MMP13和TGF-βmRNA和蛋白的表达量均降低;VEGFA、MMP13和TGF-β在联合组中蛋白着色变浅,呈弱阳性表达,联合用药组较顺铂组、当高、低组蛋白表达量降低。结论:当归贝母苦参丸加味方可以下调荷瘤小鼠肿瘤组织VEGFA、MMP13和TGF-β的表达,进而发挥抗肿瘤作用。
Objective: To study the effects of Modified Danggui Beimu Kushen Pill on the expressions of VEGFA,MMP13 and TGF-β in tumor tissues of MFC gastric cancer bearing mice. Methods: MFC gastric cancer bearing mice were employed to perform anti-tumor experiment in vivo. A total of 48 eligible mice were randomly divided into the model group,DDP group,low-dose group of Modified Danggui Beimu Kushen Pill,high-dose group of Modified Danggui Beimu Kushen Pill,DDP combined with low-dose group,and DDP combined with high-dose group. The treatment course was 14 continuous days. After the last treatment,all mice were killed to obtain complete tumor tissue for further detection of VEGFA,MMP13 and TGF-β with the method of RT-q PCR and immunohistochemistry. Results: RT-q PCR detection showed that the expressions of VEGFA,MMP13 and TGF-β mRNA in each medication group were decreased compared to those in the model group( P〈 0. 05). Immunohistochemical detection showed that the expressions of VEGFA,MMP13 and TGF-β were less colored and with weakly positive expression in the combined group,compared to DDP group and Chinese medication groups. Conclusion: Modified Danggui Beimu Kushen Pill can down-regulate the expressions of VEGFA,MMP13 and TGF-β of tumor tissue in MFC gastric cancer bearing mice to play anti-tumor role.
