摘要:目的基于脾主运化理论探讨运脾消积法治疗厌食症幼龄大鼠的代谢组学。方法基于病因模拟法建造小儿厌食症模型,通过记录摄食量和体质量的变化评价模型。60只大鼠随机分为正常组,模型组,阳性对照组(0.45 g/kg消食片),小儿开胃增食合剂高、低剂量组(11.4、5.7 g/kg)。灌胃给药42 d后收集24 h尿液,LC-MS分析尿液中代谢产物,并进行主成分分析(PCA)和正交偏最小二乘法-判别分析(OPLS-DA);通过比较质荷比搜索在线数据库Metlin,对代谢差异物进行定性。利用Metabolo Analyst 3.0软件进行代谢通路分析。结果小儿厌食症模型大鼠中有29个代谢物发生了变化,涉及9条代谢通路;经过药物干预后,与模型组相比,小儿开胃增食合剂低、高剂量组大鼠分别有29、48个代谢物发生了变化,所涉及的代谢通路主要为非饱和脂肪酸和脂肪酸生物合成、甘油磷脂代谢、苯丙氨酸代谢。结论经小儿开胃增食合剂干预后,小儿厌食症大鼠代谢失调可以发生恢复性和保护性变化,主要涉及脂质代谢途径和氨基酸代谢途径。
AIM To explore the metabolites of Dissolving Accumulation via Invigorating the Spleen Functioning Method in managing young anorexia rat models based on theory of the Spleen Governing Transformation and Transportation of Nutrients in Digestion.METHODS The infantile anorexia models established based on the etiological simulation method were evaluated in terms of the recorded changes in food intake and body weight.Sixty rats were randomly assigned into normal group,model group,positive control group(0.45 g/kg Xiaoshi Tablets),and groups of high and low dose Children’s Appetizer Food Mixture(11.4 g/kg,5.7 g/kg).The 24 hours urine collected following 42 days intragastric administration,had the metabolites analyzed by LC-MS and principal component analysis(PCA)and orthogonal partial least squares-discriminant analysis(OPLS-DA);the metabolites differentiated with reference to the mass-to-charge ratio m/z online database Metlin;and the metabolic pathway analysis performed using Metabolo Analyst 3.0 software as well.RESULTS There was a change of 29 metabolites in the anorexia rat models,involving 9 metabolic pathways.When compared with the model group,groups of Children’s Appetizer Food Mixture shared 29 and 48 metabolites changes under low dose administration and high dose administration,respectively.The metabolic pathways of non-saturated fatty acid and fatty acid biosynthesis,glycerophospholipid metabolism,and phenylalanine metabolism were mainly involved.CONCLUSION Upon metabolic disorders occuring in rat models of infantile anorexia,the intervention of Children’s Appetizer Food Mixture,can bring forth the metabolic recovery and protection via regulations of both lipid and amino acid metabolism pathway.
