摘要:根际土壤真菌群落包括生防真菌和病原真菌,研究其组成和功能有助于揭示植物病害的发生机制,为连作障碍的绿色防控提供新思路。本研究以连作当归为对象,设置水杨酸组(S.RH)、茉莉酸甲酯组(M.RH)、复合菌剂组(DK.RH)、对照组(C.RH)4个处理组,研究其叶面喷施对根际真菌群落及当归病害的影响。采用常规方法统计病害、产量,高效液相色谱串联质谱检测生长素、细胞分裂素、赤霉素、脱落酸、乙烯、褪黑素、茉莉酸、水杨酸等植物内源激素含量,Illumina Miseq高通量检测根际土壤真菌多样性,分析真菌群落组成、多样性、物种差异、功能及相关性预测。结果表明,水杨酸和茉莉酸甲酯处理均降低了根腐病发生率和产量。较对照,茉莉酸甲酯处理,根腐病发病率下降43.6%,平均百株重下降17.4%;水杨酸处理,根腐病发病率下降46.2%,平均百株重下降20.6%。复合菌剂处理降低了植物内源水杨酸(salicylic acid,SA)、茉莉酸(jasmonic acid,JA)含量,提高了脱落酸(abscisic acid,ABA)含量。水杨酸甲酯葡萄糖苷(2-Methoxycarbonylphenyl beta-D-glucopyranoside,MeSAG)的log2FC值为负无穷(-Inf),12-羟基茉莉酸(12-Hydroxyjasmonic acid,12-OH-JA)、茉莉酸异亮氨酸(jasmonoyl-L-isoleucine,JA-ILE)、茉莉酸的log2FC值分别为-1.25、-2.02、-2.10,脱落酸、脱落醛(abscisic aldehyde,ABA-ald)、脱落酸葡萄糖酯(ABA-glucosyl ester,ABA-GE)的log2FC值分别为1.49、-Inf、1.03。水杨酸和茉莉酸甲酯处理促进了木霉菌的富集,此外,水杨酸处理还促进了菌根真菌的富集;复合菌剂处理根际土壤富集了镰刀菌属、赤衣菌属真菌;对照根际土壤主要富集了烟曲霉和数丝顶多毛孢。综上,叶面喷施可以通过调控内源激素水平,尤其是脱落酸与水杨酸、茉莉酸的平衡,进而影响根际土微生物群落组成和病害发生。
The rhizosphere soil fungal community comprises both biocontrol fungi and pathogenic fungi.Understanding its composition and functions helps reveal the mechanisms underlying plant diseases and provides new insights for the green control of continuous cropping obstacles.In this study,continuous cropping of Angelica sinensis was used as the research subject,with four treatments:salicylic acid group(S.RH),methyl jasmonate group(M.RH),composite microbial agent group(DK.RH),and control group(C.RH).Disease incidence and yield were assessed with conventional methods.The contents of endogenous plant hormones,including auxins,cytokinins,gibberellins,abscisic acid,ethylene,melatonin,jasmonic acid,and salicylic acid,were detected via high-performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS).The diversity of rhizosphere soil fungi was evaluated using Illumina Miseq high-throughput sequencing,and the composition,diversity,species differences,function and correlation prediction of soil fungal communities were analyzed.The results showed that both salicylic acid and methyl jasmonate treatments reduced the incidence of root rot and yield.Compared with the control,the methyl jasmonate treatment decreased root rot incidence by 43.6%and the average weight per hundred plant individuals by 17.4%,while the salicylic acid treatment reduced root rot incidence by 46.2%and the average weight per hundred plant individuals by 20.6%.The composite microbial agent treatment reduced the endogenous levels of salicylic acid and jasmonic acid in plants,while increased the abscisic acid content.The log2FC value of methyl salicylate glucoside was negative infinity(-Inf).The log2-FC values for 12-hydroxyjasmonic acid,jasmonoyl isoleucine,and jasmonic acid were-1.25,-2.02,and-2.10,respectively,whereas those for abscisic acid,abscisic aldehyde,and abscisic acid glucosyl ester were 1.49,-Inf,and 1.03,respectively.Both salicylic acid and methyl jasmonate treatments promoted the enrichment of Trichoderma fungi,with salicylic acid additionally enhancing the enrichment of mycorrhizal fungi.In contrast,the composite microbial agent treatment led to the enrichment of Fusarium and Rhizoctonia fungi in the rhizosphere soil,whereas the control group primarily exhibited enrichment of Aspergillus fumigatus and Cladosporium cladosporioides.In conclusion,foliar spraying can modulate the composition of rhizosphere microbial communities and disease incidence by regulating endogenous hormone levels,particularly through altering the balance among abscisic acid,salicylic acid,and jasmonic acid.
