摘要:目的探讨清热逐风合剂(QZM)通过调控HIF-1α/VEGF信号通路干预胶原诱导关节炎大鼠滑膜血管新生作用机制。方法实验大鼠分为正常对照(Control)组、胶原诱导模型(CIA)组、清热逐风(QZM)组、甲氨蝶呤(MTX)组、雷公藤多苷片(TWP)组。检测大鼠的足趾肿胀度;计算关节炎指数;采用HE染色检测大鼠踝关节滑膜组织病理改变;采用ELISA检测IL-6、TNF-α、TGF-β水平,Western blot与免疫组织化学技术检测滑膜HIF-1α、VEGF蛋白的表达。结果踝关节病理切片显示QZM可以抑制关节内滑膜组织的增生,延缓滑膜血管扩张及新生。与CIA组相比,QZM、MTX及TWP组能够明显降低足趾肿胀度及关节炎指数,降低大鼠血清中IL-6、TNF-α水平(P<0.01),升高TGF-β水平(P<0.01),且能显著降低滑膜组织中HIF-1α、VEGF蛋白表达水平(P<0.01)。结论QZM调控HIF-1α/VEGF信号通路,抑制HIF-1α、VEGF蛋白的表达,减轻RA大鼠炎症水平调和抑制血管新生,从而发挥抗类风湿关节炎的作用。
Objective To investigate the mechanism of Qingrezhufeng mixture(QZM)on collagen-induced arthritis rats with synovial angiogenesis by regulating the HIF-1α/VEGF signaling pathway.Methods The experiment was divided into the control group,the collagen-induced arthritis(CIA)group,the Qingre Zhufeng(QZM)group,the methotrexate(MTX)group,and the tripterygium wilfordii polyglycoside(TWP)group.The toe swelling of rats was detected and the arthritis indexes were calculated;the pathological changes of rat ankle synovial tissue were detected by HE staining;the levels of IL-6,TNF-α,and TGF-β were detected by ELISA;the expression of synovial HIF-1α and VEGF proteins was detected by Western blot(WB)and immunohistochemistry.Results The pathological sections of ankle joint showed that QZM could inhibit the proliferation of synovial tissue in the joint and delay the expansion and angiogenesis of synovial vessels.Compared with the CIA group,the QZM,MTX and TWP groups could significantly reduce the toe swelling and arthritis indexes,reduce the levels of IL-6 and TNF-α in serum of rats(P<0.01),increase TGF-β(P<0.01),and significantly reduce the expression of synovial HIF-1α and VEGF proteins(P<0.01).Conclusion QZM regulates the HIF-1α/VEGF signaling pathway,inhibits HIF-1α and the expression of VEGF protein,reduces the inflammation levels and inhibits angiogenesis in RA rats,thereby exerting its anti-RA effects.
