摘要:目的筛选富集肉苁蓉中苯乙醇苷的最佳树脂和工艺条件,使分离纯化工艺达到最优化。方法选择7种大孔吸附树脂(AB-8、DM301、DM130、D1400、DA-201、HPD-100、D-101-I),通过树脂对苯乙醇苷的静态吸附和解析能力,筛选出最佳树脂,再通过树脂的静态特性和动态特性的筛选优化工艺。结果 DA-201的吸附解析最佳条件为:上样流速为1.5 mL.min-1,洗脱剂为90%的乙醇。按最佳工艺苯乙醇苷的纯度由25.1%上升为51.2%。结论该法简便、可行,为肉苁蓉中苯乙醇苷的纯化提供了参考。
OBJECTIVE To investigate the optimal macroporous adsorption resin and process to separate phenylethanoid glycosides from Cistanche tubulosa.METHODS Seven types of macroporous adsorption resin were evaluated for separating efficiency by measuring the adsorption ratio,eluting ratio of phenylethanoid glycosides.RESULTS The optimal technologies of DA-201 macroprous dosorption resin were as follows: the simple flow was 1.5 mL·min-1,the column was washed by 95% ethanol.After enrichment and purification,the product purity of phenylethanoid glycosides was up to 51.2%.CONCLUSION This method is simple,feasible and can provide useful reference for the isolation and purification of phenylethanoid glycosides from Cistanche tubulosa.
