摘要:目的观察小儿平喘方对哮喘模型大鼠血清中γ-干扰素(IFN-γ)、白细胞介素-4(IL-4)含量、IFN-γ/IL-4比值的影响,从而探讨小儿平喘方治疗支气管哮喘的药效机制。方法选取SD大鼠32只,随机分为4组:正常对照组、哮喘模型组、小儿平喘方小剂量组(小剂量组)和小儿平喘方大剂量组(大剂量组),每组8只,采用卵清蛋白(OVA)致敏和雾化吸入激发制备大鼠哮喘模型,用双位点夹心酶联免疫吸附试验测定各组大鼠血清中IL-4和IFN-γ含量。结果与正常对照组比较,哮喘模型组大鼠血清IFN-γ的含量与IFN-γ/IL-4比值均降低(P<0.05),而IL-4的含量则显著升高(P<0.05);与哮喘模型组比较,大、小剂量组大鼠血清IFN-γ的含量与IFN-γ/IL-4比值均显著升高(P<0.01),而IL-4的含量则降低(P<0.05)。结论小儿平喘方能显著降低支气管哮喘模型大鼠血清IL-4的含量,提高IFN-γ的含量及失衡的IFN-γ/IL-4比值,从而调节免疫功能,减轻炎症反应,控制哮喘的发作。
Objective To observe the influence of Xiaoer Pingchuan Fang on IL-4 and IFN-γ levels and the IFN-γ/IL-4 ratio of the serum in asthmatic rats, so as to investigate the pharmacodynamic mechanism of Xiaoer Pingchuan Fang in treating children bronchial asthma. Methods 32 SD rats were randomly divided into 4 groups : a normal control group, an asthma model group, a Xiaoer Pingchuan Fang group of low dose and a Xiaoer Pingchuan Fang group of large dose, 8 each. The ovalbumin (OVA) sensitization and the atomization inhalation were used to motivate and prepare for models of asthma rats, with the use of double loci enzyme combined with the immunosorbent experiment to determine the IL-4 and IFN-γcontent of the rat serum in each group. Results Compared with those in the normal control group, the IFN-γ content and the IFN-γ /IL-4 ratio in the asthma model group were both lowered(P〈0.05 ), while the content of IL-4 increased(P〈0.05). Compared with those in the asthma model group, both the IFN-γ content and the IFN-γ/IL-4 ratio in the Xiaoer Pingchuan Fang group of low dose and the Xiaoer Pingchuan Fang group of large dose significantly increased(P〈0.01 ) , but the content of IL-4 increased( P 〈 0. 05 ). Conclusion Xiaoer Pingchuan Fang can significantly reduce the content of IL-4 in bronchial asthma rats serum and increase the content of IFN-γ and imbalanced ratio of IFN-γ IL-4, accordingly regulating the immune function, reducing inflammation reactions and controlling the asthma attacks.
