摘要:目的:通过正交试验法优选甘肃米仓红芪微波炮制工艺,分析微波炮制法对米仓红芪中黄酮类成分含量的影响,建立红芪微波炮制方法,为红芪炮制工业生产规范化提供科学依据。方法:采用甲醇回流提取红芪中毛蕊异黄酮和芒柄花素,用高效液相色谱法进行毛蕊异黄酮和芒柄花素的测定含量,采用L_9(3~4)正交试验法,优选红芪的最佳微波炮制工艺。结果:优选出的最佳微波条件是:温度低火,时间5 min,厚度2 cm,其炮制品毛蕊异黄酮含量为3.984μg·g^(-1),芒柄花素含量为42.314μg·g^(-1),水分6.66%,灰分1.34%,醇溶出度41.98%。结论:结果表明,红芪微波炮制品能够达到2015年版《中国药典》要求,且方法简单可行,重现性好,可作为甘肃道地药材红芪炮制品质量控制的有效方法,本实验为进一步研究红芪最佳炮制工艺提供科学依据。
OBJECTIVE To optimize the best microwave processing method for Hedysari by orthogonal test,analyze the influence of processing method on the contents of flavonoids,establish the microwave processing method,and provide a scientific basis for standardized processing and industrialization of Hedysari.METHODS Methanol reflux was used to extract and high efficiency liquid chromatography method was used to determine calycosin and formononetin.The best microwave processing method for Hedysari was optimized by using L_9(3~4) orthogonal test.RESULTS The optimum microwave conditions optimized:low temperature fire for 5 min,thickness 2 cm,calycosin 3.984 g·g-1,formononetin 42.314 g·g-1,moisture 6.66%,ash 1.34%,alcohol dissolution 41.98%.CONCLUSION The results indicate that the microwave processed Radix Hedysari can satisfy the requirements in 2015 edition of China Pharmacopoeia.The method is simple,feasible and reproducible,and can be used as an effective method for quality control of Radix Hedysari in Gansu province.This experiment provides scientific evidence for further investigate the best processing method for Radix Hedysari.
