摘要:目的探讨当归红芪超滤物(RAS-RH)对阿霉素致心力衰竭大鼠的炎症因子及PI3K、Akt蛋白的影响。方法将60只Wistar雄性大鼠随机取12只为空白组后,剩余大鼠均采用腹腔注射阿霉素方法诱导心力衰竭大鼠模型。造模4周末后采用多普勒超声心动图机测定大鼠心功能以鉴定模型,造模成功后将大鼠随机分为模型组、西药组(盐酸贝那普利1.0mg·kg^(-1)·d^(-1))、中药组(RAS-RH 0.3mg·kg^(-1)·d^(-1))、中西药联合组(盐酸贝那普利1.0mg·kg^(-1)·d^(-1)+RAS-RH 0.3mg·kg^(-1)·d^(-1))。对药物组进行药物干预6周末后采用多普勒超声心动图机测定大鼠心功能,ELISA法检测血清TNF-α、IL-6、IL-β、CRP的表达量,HE染色法观察大鼠心肌组织病理形态学变化,Western Blot法检测心肌组织中PI3K、Akt蛋白的表达量。结果与空白组相比,模型组大鼠心功能降低(P<0.05);与模型组相比,西药组、中西药联合组给药后大鼠心功能均改善(P<0.05)。与空白组相比,模型组大鼠TNF-α、IL-6、IL-β、CRP等炎症因子及PI3K、Akt蛋白的表达均异常(P<0.05);与模型组比较西药组、中药组、中西药联合组大鼠TNF-α、IL-6、IL-β、CRP等炎症因子及PI3K、Akt蛋白的表达量显著改善(P<0.05)。结论当归红芪超滤物对阿霉素致心力衰竭大鼠血清TNF-α、IL-6、IL-β、CRP等炎症因子及心肌组织PI3K、Akt蛋白的表达具有改善作用。
Objective To investigate the effects of Radix Angelica Sinensis and Radix Hedysari Ultrafiltration(RAS-RH) on inflammatory factors,PI3 K and Akt proteins in rats with adriamycin-induced heart failure.Methods After randomly selecting 12 of 60 Wistar male rats as the blank group,the remaining rats were injected intraperitoneally with adriamycin to induce a rat model of heart failure.After 4 weeks of modeling,a Doppler echocardiography machine was used to determine the heart function of the rat to identify the model.After the modeling was successful,the rats were randomly divided into a model group and a western medicine group(benazepril hydrochloride 1.0 mg·kg^(-1)),Chinese medicine group(RAS-RH 0.3 mg·kg^(-1)),Chinese and western medicine combination group(Benazepril hydrochloride 1.0 mg·kg^(-1)+ RAS-RH 0.3 mg·kg^(-1)).After 6 weeks of drug intervention in the drug group,the heart function of rats was measured by Doppler echocardiography,the expression of serum TNF-α,IL-6,IL-β,and CRP was detected by ELISA,and the rats were observed by HE staining method.Pathological changes of myocardial tissue.Western Blot method was used to detect the expression of PI3 K and Akt protein in myocardial tissue.Results Compared with the blank group,the heart function of rats in the model group was reduced(P<0.05);compared with the model group,the heart function of the rats in the western medicine group and the combination group of Chinese and Western medicine improved after administration(P<0.05).Compared with the blank group,the expression of inflammatory factors such as TNF-α,IL-6,IL-β,CRP and PI3 K and Akt protein in the model group were abnormal,and the difference was statistically significant(P<0.05);the western medicine group,The expression of inflammatory factors such as TNF-α,IL-6,IL-β,CRP,PI3 K and Akt protein in the traditional Chinese medicine group and the combination group of Chinese and Western medicine were significantly improved,and the difference was statistically significant(P<0.05).Conclusion Radix Angelica Sinensis and Radix Hedysari Ultrafiltration can improve serum TNF-α,IL-6,IL-β,CRP and other inflammatory factors and the expression of PI3 K and Akt protein in myocardial tissue in rats with adriamycin-induced heart failure.
