摘要:目的:探讨基于iTRAQ技术分析小儿开胃增食合剂干预下厌食症大鼠胃窦组织中蛋白质组学变化及临床意义。方法:构建SD幼龄大鼠厌食症模型,提取胃窦组织总蛋白,用iTRAQ试剂标记后进行质谱检测并分析差异性表达的蛋白质。对差异表达的蛋白质进行GO富集分析、KEGG富集分析。收集小儿厌食症患者、小儿开胃增食合剂治愈的患者及同期参与体检的健康小儿全血提取总蛋白。用Western Blotting法检测厌食症大鼠全血和小儿厌食症患者全血中差异蛋白的相对表达量。结果:与空白对照组相比较,模型对照组大鼠Scamp3、Tmem86a和Akr1b7蛋白的相对表达量明显上调(P<0.05),Smarcc1、Nudt13、Rpp30、Rps4y2、Art2b和Glmp蛋白的相对表达量明显下调(P<0.05);与模型对照组相比较,小儿开胃增食合剂14.4 g/kg组大鼠Scamp3、Tmem86a和Akr1b7蛋白的相对表达量明显下调(P<0.05),Smarcc1、Nudt13、Rpp30、Rps4y2、Art2b和Glmp蛋白的相对表达量明显上调(P<0.05)。与正常对照组比较,小儿厌食症组中Scamp3和Tmem86a蛋白的相对表达量明显上调(P<0.05),Smarcc1、Nudt13、Rpp30和Glmp蛋白的相对表达量明显下调(P<0.05);与小儿厌食症组比较,小儿开胃增食合剂14.4 g/kg组中Scamp3和Tmem86a蛋白的相对表达量明显下调(P<0.05),Smarcc1、Nudt13、Rpp30和Glmp蛋白的相对表达量明显上调(P<0.05)。结论:小儿开胃增食合剂干预下厌食症大鼠胃窦组织及全血中差异蛋白组学的改变与小儿厌食症患者全血蛋白的差异表达具有一定的相关性,本研究为小儿开胃增食合剂治疗厌食症的临床应用提供了一定的理论支持。
Objective: To investigate the proteomics and clinical significance of gastric antrum in rats with anorexia under the intervention of Xiaoer Kaiwei Zengshi Mixture based on iTRAQ technology. Methods: SD juvenile rat anorexia model was established, the total protein from the gastric antrum tissue was extracted and labeled with iTRAQ reagent, then tested by mass spectrum to analyze the differentially expressed proteins. GO enrichment analysis and KEGG enrichment analysis were performed on the differentially expressed proteins. Whole blood was collected from children with anorexia, children cured with Xiaoer Kaiwei Zengshi mixture, and healthy children who participated in physical examinations to extract the total protein. Western Blotting was used to detect the relative expressions of differential proteins in the whole blood of anorexia rats and the whole blood of children with anorexia. Results: Compared with the control group, the relative expressions of Scamp3, Tmem86 a and Akr1 b7 protein in the model group were significantly up regulated(P<0.05), and the relative expressions of Smarcc1, Nudt13, Rpp30, Rps4 y2, Art2 b, and Glmp were significantly down regulated(P<0.05). Compared with the model group, the relative expressions of Scamp3, Tmem86 a and Akr1 b7 proteins in 14.4 g/kg Xiaoer Kaiwei Zengshi mixture group were significantly down regulated(P<0.05), and the relative expressions of Smarcc1, Nudt13, Rpp30, Rps4 y2, Art2 b and Glmp protein were significantly up regulated(P<0.05). Compared with the control group, the relative expressions of Scamp3 and Tmem86 a protein in the infantile anorexia group were significantly up regualted(P<0.05), and the relative expressions of Smarcc1, Nudt13, Rpp30 and Glmp protein were significantly down regulated(P<0.05). Compared with the children with anorexia group, the relative expressions of Scamp3 and Tmem86 a protein in 14.4 g/kg Xiaoer Kaiwei Zengshi Mixture group were significantly down regulated(P<0.05), and the relative expressions of Smarcc1, Nudt13, Rpp30 and Glmp protein were significantly up regulated(P<0.05).Conclusion: The change of differential proteomics in gastric antrum tissue and whole blood of anorexic rats under the intervention of Xiaoer Kaiwei Zengshi Mixture is related to the differential expression of whole blood protein in children with anorexia. This study provides certain theoretical support for the clinical effect of Xiaoer Kaiwei Zengshi mixture in the treatment of anorexia.
