摘要:目的建立一条适合于工业化生产提取甘草并超滤提取液的工艺路线。方法采用响应面法,以提取时间、提取次数、氨水浓度为提取考察因素,采用L9(3~4)正交设计表,以膜面错流速度、操作压力、料液温度为超滤纯化考察因素,以甘草酸、甘草苷的含量(保留率)及出膏率(除杂率)为评价指标,优选甘草氨水提取及陶瓷膜超滤的最佳工艺参数。结果最佳提取工艺为:提取时间3 h、提取次数2次、氨水浓度0.70%、料液比1:20;最佳超滤工艺为陶瓷膜孔径50 nm、膜面错流速度5.3 m·s^(-1)、压力0.12 MPa、料液温度25℃。在上述工艺下,甘草酸、甘草苷的保留率达95.36%、95.43%,除杂率达18.13%。结论所用方法简便可行、生产成本低,有良好的工业化生产前景。
OBJECTIVE To establish a suitable process for industrial extraction and ultrafiltration of Glycyrrhizae Radix et Rhizoma extract.METHODS Response surface method was used with extraction time,extraction times and ammonia concentration as extraction factors.L9(34)orthogonal design table was used with membrane cross-flow velocity,operating pressure and feed liquid temperature as the extraction factors.The contents of glycyrrhizic acid and liquiritin(retention rate)and extract rate(impurity removal rate)as evaluation indexes to optimize the extraction and ultrafiltration process parameters of Glycyrrhizae Radix et Rhizoma.RESULTS The optimal extraction process was 3 h of extraction time 2 times,0.70%ammonia concentration,solid-liquid ratio of 1∶20.The optimal ultrafiltration process was as follows:the pore size of the ceramic membrane was 50 nm;the cross-flow velocity of the membrane surface was 5.3 m·s^(-1);the pressure was 0.12 MPa;and the liquid temperature was 25℃.The retention rate and impurity removal rate of glycyrrhizic acid and liquiritin were 95.36%,95.43%and 18.13%,respectively.CONCLUSION The method is simple,feasible and of low production cost,and it has good industrial production prospect.
