摘要:目的基于网络药理学分析秦艽-威灵仙药对治疗类风湿关节炎(RA)的作用机制。方法通过TTD和DrugBank数据库检索与RA相关的疾病靶点;基于中药系统药理学分析平台(TCMSP)数据库检索秦艽-威灵仙药对的成分,并以生物口服利用度(OB)≥30%和类药性(DL)≥0.18为标准筛选活性成分;运用Swiss Target Prediction数据库检索各活性成分的作用靶点,通过映射筛得治疗RA的重要靶点并构建秦艽-威灵仙药对的“活性成分-重要靶点”网络以筛选重要成分和核心靶点;将核心靶点通过STRING数据库建立蛋白互作网络图;利用DAVID平台进行KEGG信号通路及GO生物过程富集以整理重要通路和重要生物过程。结果通过筛选共挖掘出秦艽-威灵仙药对活性成分13个、重要靶点30个、核心靶点12个、生物过程3条及重要信号通路8条。结论初步揭示了秦艽-威灵仙药对治疗RA的药效物质基础,为秦艽-威灵仙药对治疗RA的临床应用及实验研究提供了参考依据。
Objective To analyze the action mechanism of Gentiana macrophylla Pall.and Clematis chinensis Osbeck couplet medicinals in the treatment of rheumatoid arthritis(RA)based on network pharmacology.Methods The target in diseases related to RA was searched by TTD and DrugBank databases.Components of Gentiana macrophylla Pall.and Clematis chinensis Osbeck couplet medicinals were detected based on Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).The main active ingredients were screened by oral bioavailability(OB)≥30%and drug-like(DL)≥0.18 and the Swiss Target Prediction system was used to retrieve the corresponding targets of each active ingredient,through mapping and screening the important targets of RA treatment,and constructing the network of“active components important targets”of Gentiana macrophylla Pall.and Clematis chinensis Osbeck couplet medicinals to screen the important components and key targets.The protein interaction network diagram was set up through key targets of the construction of STRING database.DAVID platform was used to carry out KEGG signal pathway and GO biological process enrichment,and to sort out important pathways and biological processes.Results Through screening,13 main active ingredients,30 important targets,12 key targets,3 biological processes and 8 important signaling pathways were mined from Gentiana macrophylla Pall.and Clematis chinensis Osbeck couplet medicinals.Conclusion The material basis of pharmacodynamics of Gentiana macrophylla Pall.and Clematis chinensis Osbeck couplet medicinals is revealed preliminarily,which provides a reference for clinical application and experimental study of it.
