摘要:【目的】探讨^(60)Coγ射线对大鼠血清性激素水平及PI3K/Akt/Foxo3a信号通路蛋白表达的影响,明确辐射对卵巢组织损伤的分子机制.【方法】通过一次6.0 Gy^(60)Coγ射线对大鼠进行照射后,分别在第3、7、14、21天,通过TUNEL法观察卵巢组织细胞凋亡情况、血清LH、FSH和雌激素(E2)水平及免疫组化法观察卵巢组织Akt、磷酸化Akt、Foxo3a、磷酸化Foxo3a蛋白水平的表达.【结果】辐射损伤造成卵巢颗粒细胞明显凋亡(P<0.01);而且随着辐射时间的延长细胞凋亡数量逐渐增加,在辐射第21天达到高峰(P<0.01);大鼠血清FSH、LH和E2水平随着辐射对卵巢功能的损伤而逐渐降低(P<0.01);卵巢组织蛋白测定总Akt,p-Akt、Foxo3a蛋白的表达呈现逐渐增加趋势(P<0.01),尤以总Akt和Foxo3a蛋白的表达在辐射第21天最显著(P<0.01);而p-Foxo3a蛋白表达逐渐下降(P<0.01),尤以辐射第21天最显著(P<0.01).【结论】辐射导致大鼠POF的原因,一方面激活PI3K/Akt/Foxo3a信号通路,导致颗粒细胞以及卵母细胞凋亡,另一方面垂体腺细胞凋亡影响促性腺激素分泌.
【Objective】To investigate the effects of^(60)Coγ-ray on the levels of serum sex hormone and protein expression in PI3K/Akt/Foxo3a signaling pathway in rats and elucidate the molecular mechanism of radiation damage to ovarian tissue.【Method】The apoptosis of ovarian tissue cells was observed by TUNEL method and serum LH,FSH and estrogen(E2)hormone levels were detected,respectively at 3,7,14 and 21 days after one-time irradiation by 6.0 Gy^(60)Coγ-ray.The expression levels of Akt,phosphorylated Akt,FoxO3a and phosphorylated FoxO3a proteins in ovarian tissue were determined by the way of immunohistochemistry.【Result】The distinct ovarian-cell apoptosis occurred after the irradiation(P<0.01),and the number of apoptotic cells increased with the extension of radiation time and reached a peak at 21 d after irradiation(P<0.01).The levels of serum FSH,LH and E2 in rats gradually decreased with the injury of ovarian function caused by radiation(P<0.01).With the extension of irradiation time,the expression of total Akt,p-Akt and Foxa3a proteins gradually increased(P<0.01),among which the expression of total Akt and Foxa3a proteins was the most significant at the 21 d after the radiation(P<0.01).However,the expression of Foxa3a protein decreased gradually(P<0.01),which was the most significant at the 21d after the radiation(P<0.01).【Conclusion】The causes of POF in rats induced by radiation were that PI3K/Akt/Foxo3a signaling pathway was activated,which led to the apoptosis of granulosa cells and oocytes,and that the secretion of gonadotropin was affected due to the apoptosis of pituitary adenosine cells.
