摘要:目的研究红芪总黄酮对人早幼粒白血病HL-60细胞诱导分化的作用及其分子机制。方法HL-60细胞经红芪总黄酮处理后,检测细胞NBT还原能力的改变,用流式细胞仪测定细胞增殖周期及CD11b、C-fos表达。结果红芪总黄酮作用于HL-60细胞后,NBT还原能力增强;CD11b表达升高;细胞被阻滞于G0/G1及G2/M期,S期细胞数相应减少;C-fos基因表达增强。结论红芪总黄酮可诱导HL-60细胞分化成熟,其机制可能与上调C-fos基因表达有关。
Objective: To investigate the effect and mechanism of total flavonoids of Hedysarum polybotry on induction of differentiation in human leukemia HL-60 cells. Method After the treatment of HL-60 cells with total flavonoids of Hedysarum polybotry, the cell differentiation was detected with NBT reduction method. Cell cycle, CD11b and C-los were analysed by the flow cytometry. Result The positive rate of NBT reduction and the expression of CD11b were significantly increased. Similar, the expression of C-los gene was upregulated. The growth of HL-60 cells was arrested at G0/G1 and G2/M phase. Conclusion Total flavonoids of Hedysarum polybotry could induce differentiation of HL-60 cells. Its molecular mechanism might be related to the modulation of gene expressions associated with the proliferation and differentiation, which leads to the inhibition of DNA synthesis.
