摘要:目的:探讨平胃胶囊治疗肝郁脾虚型功能性消化不良的分子机制。方法:夹尾刺激、饮食失节、过度疲劳的复合因素方法建立肝郁脾虚型FD大鼠模型,造模10d,将16只造模成功的大鼠随机分为模型组和平胃胶囊组,每组各8只,平胃胶囊组以0.24g/mL的混悬液1mL/100g灌胃3周,正常组和模型组以1mL/100g生理盐水灌胃,3组均灌胃给药3周,2次/d。记录大鼠体质量、饮食,测定胃排空及肠推进,胃组织HE染色,蛋白质组学方法筛选差异表达的蛋白,采用MALDI—TOF—MS质谱仪进行质谱分析。结果:平胃胶囊组大鼠体质量、饮食均较模型组上升(P<0.05);经典夹尾刺激、配合不规律饮食、过度疲劳的方法可成功复制肝郁脾虚型FD大鼠模型;平胃胶囊具有增加胃排空和肠推进,促进胃肠动力的作用;成功鉴定出10种蛋白,模型组与正常组相比有差异而平胃胶囊可使差异趋势逆转的蛋白点为8个,其中5个表达上调,3个表达下调。结论:平胃胶囊治疗肝郁脾虚型FD的分子机制可能与改善细胞骨架和形态、能量代谢、生物氧化等有关。
Objective:To explore the possible molecular mechanism of Pingwei Capsule in treating functional dyspepsia with liver depression and spleen deficiency type. Methods:The FD rat model of liver depression and spleen deficiency type was established by compound factors including tail stimulation,diet loss and fatigue for 10 days. Sixteen successful models were randomly divided into model group and Pingwei Capsule group,8 rats in each group. Pingwei Capsule group was given 0.24 g/mL suspension for 3 weeks,normal group and model group were given 1 mL/100 g normal saline for 3 weeks,twice a day. Weight and diet of rats were recorded,gastric emptying and intestinal propulsion were measured,HE staining of gastric tissues was performed,proteomics methods were used to screen differentially expressed proteins,and MALDI-TOF-MS mass spectrometry was used to analyze the proteins. Results:The weight and diet of rats in Pingwei Capsule group were higher than model group. The FD rat model of liver depression and spleen deficiency type can be successfully reproduced by classical tail clamping stimulation,irregular diet and excessive fatigue. Pingwei Capsule can increased gastric emptying and intestinal propulsion,and promote gastrointestinal motility. Ten proteins were identified successfully. There were 8 protein spots in the model group which were different from the normal group. PingweiCapsule could reverse the trend of the difference. Five of them were up-regulated and three were downregulated. Conclusion:The molecular mechanism of Pingwei Capsule in treating FD of liver depression and spleen deficiency type may be related to the improvement of cytoskeleton and morphology,energy metabolism and biological oxidation.
