摘要:目的:探讨地龙降压胶囊对自发性高血压大鼠(SHR)血压的影响机制。方法:将40只SHR随机分为5组,包括模型对照组、地龙降压胶囊0.126 g/kg、0.252 g/kg、0.504 g/kg组、贝那普利0.9 mg/kg组;取8 w龄雄性魏-凯二氏大鼠(WKY)8只作为空白对照组,连续灌胃给予相应药物或生理盐水9 w。每周测大鼠血压,并于给药结束后采血检测血清肾素(Renin)、血管紧张素Ⅱ(Angiotensin II, AngⅡ)、醛固酮(Aldosterone, Ald)水平变化情况;分离胸主动脉,逆转录聚合酶链反应(RT-PCR)检测血管紧张素Ⅱ1型受体(AT1R)mRNA的表达,蛋白印迹法测定转化生长因子-β1(TGF-β1)、血管内皮生长因子(VEGF)蛋白表达。结果:与空白对照组相比,模型对照组收缩压、舒张压明显升高,而且随着观测时间增加血压升高更加明显;同时模型对照组动物血清中Renin活性、AngⅡ、Ald水平及AT1R mRNA、TGF-β1、VEGF蛋白表达均明显升高(P<0.05)。与模型对照组相比,地龙降压胶囊0.252、0.504 g/kg组、贝那普利0.9 mg/kg组在给药干预后收缩压分别在第3 w、第3 w、第1 w出现明显下降;同时地龙降压胶囊0.252、0.504 g/kg组、贝那普利0.9 mg/kg组动物血清中Renin、AngⅡ、Ald水平及AT1R mRNA、TGF-β1、VEGF蛋白表达均明显降低(P<0.05)。结论:地龙降压胶囊可能通过抑制RAAS系统过度激活及TGF-β1、VEGF等蛋白的表达从而产生明显的降压作用。
Objective: To explore the effect of Dilongjiangya Capsule on blood pressure in spontaneously hypertensive rats(SHR) and its mechanism. Methods: 40 SHR were randomly divided into 5 groups including the model group, Dilongjiangya Capsule groups(0.126 g/kg, 0.252 g/kg group and 0.504 g/kg), benazepril 0.9 mg/kg group, while 8 male 8-week-old WKY rats were selected as the control group. Rats were orally administered with corresponding drugs or saline water for 9 weeks. Blood pressures of rats were measured weekly. After the last administration, blood was collected to detect the levels changes of serum Renin, angiotensin II(Ang II) and aldosterone(Aldosterone, Ald). Thoracic aorta was isolated, the expression of AT1 R mRNA was detected by RT-PCR. The expression of TGF-β1 and VEGF protein was determined by Western blot. Results: Compared with the control group, the systolic and diastolic blood pressure in the model group significantly increased,according with the observation time increased. At the same time, the levels of Renin, AngⅡ, Ald in the serum were increased, the AT1 R mRNA, TGF-β1, VEGF protein expressions were increased in the model group(P<0.05). Compared with the model group,after the drug intervention, blood pressure was decreased at the 3 rd week, 3 rd week and 1 st week in Dilongjiangya capsule 0.504 g/kg group, 0.252 g/kg group and benazepril 0.9 mg/kg group, respectively. Meanwhile, the serum levels of Renin, AngⅡ, Ald in Dilongjiangya capsule 0.504 g/kg group, Dilongjiangya capsule 0.252 g/kg group and benazepril 0.9 mg/kg group were decreased, the AT1 R mRNA, TGF-β1, and VEGF protein expressions were also reduced blood pressure. Conclusion: Dilongjiangya capsule has antihypertensive effect by inhibiting the excessive activation of RAAS system and the expression of TGF-β1, VEGF and other proteins.
