摘要:目的:探讨敦煌古方“紫苏煎”治疗慢性支气管炎的作用机制。方法:采用改良烟熏法复制慢性支气管炎大鼠模型,实验动物随机分为模型组、冲剂组、紫苏煎低剂量组、紫苏煎高剂量组及不加处理的空白对照组。造模各组以高、低剂量紫苏煎、止咳化痰冲剂和生理盐水进行干预,测定各组大鼠血清、肺组织中内皮素-1(ET-1)及一氧化氮(NO)的含量。结果:模型组大鼠血清、肺组织中ET-1较正常空白组明显升高(P<0.01);NO含量较正常空白组明显降低(P<0.01)。与模型组比较,紫苏煎高剂量组、紫苏煎低剂量组及冲剂组血清、肺组织中ET-1明显降低(P<0.05或P<0.01);NO含量均明显升高(P<0.05或P<0.01)。与冲剂组比较,紫苏煎高剂量组血清、肺组织中ET-1含量明显低于冲剂组(P<0.05或P<0.01);NO含量均高于冲剂组(P<0.05或P<0.01)。结论:“紫苏煎”可显著提高慢性支气管炎模型大鼠的NO含量,降低ET-1的含量。提示该方可以调节和保持血管、气管舒缩平衡,减轻炎症反应。
Objective: To investigate the internal mechanism on treating chronic bronchitis of ancient formula Zisujian of Dunhuang. Methods:Duplicated the animal model of chronic bronchitis with refined smokingfumigated method, the rats were randomly divided into model group, Chongji group, low dose group, high dose group and normal control group that didn' t treated. Made intervention with high and low dose Zisujian as well as Zhike Huatan Chongji and 0.9 % NS, determined the contents of ET-1 and NO in the serum and lung tissue. Results: The content of ET-1 in the serum and lung tissue of the model group was significantly improved than that of the normal group (P 〈 0. 01 ), and the content of NO was significantly reduced (P 〈 0. 01 ). Compared with the model group, the contents of ET-1 of high and low dose group and chongji group were significantly reduced (P 〈 0. 05 or P 〈 0. 01 ) ,but NO was opposite. Compared with Chongji group,the content of ET-1 of high dose group was significantly reduced ( P 〈 0. 05 or P 〈 0, 01 ), and NO was opposite. Conclusion : Zisujian can significantly improve the content of NO of model rats with chronic bronchitis, and reduce the content of ET-1. The formula can reduce inflammatory reaction by adjusting and keeping the balance of diastolization and contraction of blood vessel and trachea.
