摘要:目的研究清热排毒胶囊对急性痛风关节炎模型大鼠氧化应激损伤的保护作用。方法将60只大鼠随机分为空白组、模型组、双氯芬酸钠缓释片组,清热排毒胶囊低、中、高组。各组给与相应药物灌胃7天,于第5日灌胃给药1 h后采用MSU溶液右踝关节腔注射制备大鼠急性痛风性关节炎模型。各组大鼠于造模后48 h处死并取材。观察各组大鼠一般情况,测量踝关节肿胀程度,检测踝关节液中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)等氧化应激指标及踝关节软骨Nrf2蛋白表达情况。结果与模型组比较,SOD、GSH-Px活性升高,MDA含量降低(P<0.05),Nrf2蛋白表达量明显降低(P<0.01)。结论清热排毒胶囊对急性痛风性关节炎大鼠踝关节氧化应激损伤具有一定保护作用,其机制可能与抗氧化有关。
Objective To study the effect of Qingre Paidu capsule on the expression of IL-1 R and ICAM-1 in serum of rats with acute gouty arthritis. Methods Sixty rats were randomly divided into blank group, model group, diclofenac sodium sustainedrelease tablet group, and Qingre Paidui Capsule low, middle and high groups. Each group was given intragastric administration of the corresponding drug for 7 days, and on the 5 th day after intragastric administration for 1 h, an acute gouty arthritis model in rats was prepared by intraarticular injection of MSU solution. Measuring degree of swelling of ankle joint.The expression of superoxide dismutase(SOD), malondialdehyde(MDA), glutathione peroxidase(GSH-Px) and Nrf2 protein in ankle joint fluid were detected. Results Compared with the model group, SOD and GSH-Px activity increased, MDA content decreased(P<0.05), and Nrf2 protein expression decreased significantly.(P<0.01). Conclusion Qingre Paidu Capsule has protective effect on oxidative stress injury of ankle joint in rats with acute gouty arthritis, and its mechanism may be related to antioxidation.
