摘要:目的优选消风止痒方的最佳提取工艺。方法采用单因素试验考察加水量、提取时间和提取次数对提取工艺的影响,以升麻素苷、阿魏酸、5-O-甲基维斯阿米醇苷、总黄酮含量和浸膏得率为综合评价指标,以加水量和提取时间为考察因素,通过星点设计-效应面法设计试验,优选消风止痒方的提取工艺。结果根据综合评分结果,优选得到消风止痒方最佳提取工艺为:加15倍量水,提取次数2次,提取时间100 min。3批验证得综合评分平均值为98.4392,理论预测值为96.2315,偏差率为2.29%,结果与预测值相对标准偏差均小于5%。结论优选的最佳提取工艺合理、可行,且提取率高,重复性及稳定性好,可用于消风止痒方的提取。
Objective To optimize the extraction process of Xiaofeng Zhiyang Fang(消风止痒方).Methods Single factor experiment was used to investigate the effects of water amount,extraction duration and the number of extraction times on the extraction process.The content of prim-O-glucosylcimifugin,ferulic acid,5-O-methylvisammioside,total flavonoids and extract yielding rate were taken as comprehensive evaluation indexes,and water amount and extraction time were taken as investigation factors.The extraction process of Xiaofeng Zhiyang Fang was optimized by central composite design-response surface methodology.Results According to the comprehensive score results,the optimal extraction process of Xiaofeng Zhiyang Fang were as follows:adding 15-fold of water,extracting times were twice,extraction duration was 100 min.The average comprehensive score of the three batches was 98.4392,the theoretical predicted value was 96.2315,and the deviation rate was 2.29%.The relative standard deviation between the results and the predicted value was less than 5%.Conclusion The optimized extraction process is reasonable and feasible with high extraction rate,good repeatability and stability,which can be used for the extraction of Xiaofeng Zhiyang Fang.
