摘要:目的研究小苞黄芪的化学成分,并通过网络药理学初探其抗阿尔兹海默症(AD)的作用机制。方法采用硅胶柱色谱、制备色谱法对小苞黄芪乙醇提取物进行分离纯化,通过核磁共振波谱、气质联用技术鉴定其化学成分;利用网络药理学筛选出小苞黄芪抗AD的潜在活性成分及核心靶点,基于核心靶点进行生物学功能富集分析。结果从小苞黄芪乙醇提取物中共鉴定得到43个化合物,包括黄酮类、三萜类、酯类、酚酸类等。结合网络药理学从中筛选出33个潜在活性成分,主要通过调控神经退行性疾病途径、c-AMP信号通路、PI3K-Akt信号通路等来发挥抗AD的作用。结论丰富了小苞黄芪的化学成分信息,为小苞黄芪抗AD的相关研究提供了理论基础。
OBJECTIVE To study the chemical compositions of Astragalus pratti Simps and to explore the mechanism against Alzheimer's disease(AD)through network pharmacology.METHODS The ethanol extract of A.pratti was separated and purified by silica gel column chromatography and preparation chromatography.The chemical compositions were identified by NMR and CC-MS.The potential active ingredients and core targets of A.pratti.against AD were screened by Network pharmacology,then biological function enrichment analysis based on core targets.RESULTS TA total of 43 compounds were identified from the ethanol extract of A.pratti including flavonoids,triterpenoids,esters,phenolic acids and other compounds.33 potential active components were selected,which mainly exert anti-AD effects by regulating pathways such as neurodegenerative diseases,c-AMP signaling pathway,and PI3K-Akt signaling pathway.CONCLUSION This study enriches the chemical composition information of A.pratti,and provides a scientific theoretical basis for the research of A.pratti against AD.
