摘要:目的探讨慈菇消脂丸含药血清对Hh信号通路相关差异表达miRNA的调控作用。方法利用生物信息学技术筛选不同浓度慈菇消脂丸含药血清干预组差异表达miRNAs,并进行GO和KEGG富集分析,筛选出Hh信号通路相关miRNA,运用Q-PCR技术进行验证。结果生物信息学筛选差异miRNAs靶基因,结果显示:与正常组比较,2%含药血清组筛选出63个差异miRNAs,34个基因上调和29个基因下调;4%含药血清组筛选出72个差异miRNAs,37个基因上调和35个基因下调;6%含药血清组筛选出82个差异miRNAs,41个基因上调和41个基因下调。通过GO和KEGG富集分析,发现Hh信号通路中9个显著差异miRNAs:miR-149-5p、miR-199a-5p、miR-210-3p、miR-222-3p、miR-27b-3p、miR-34a-5p、miR-378a-3p、miR-497-5p、miR-671-5p,通过查阅相关文献,最终确定3个靶基因miR-378a-3p、miR-199a-5p、miR-34a-5p进行Q-PCR验证,结果显示:与正常对照组比较,不同浓度的慈菇消脂丸含药血清处理组miR-378a-3p基因表达上调(P<0.01),miR-199a-5p、miR-34a-5p基因表达下调(P<0.01)。结论中药慈菇消脂丸含药血清可以使LX2细胞中miR-199a-5p、miR-34a-5p基因表达下调,miR-378a-3p基因表达上调,可能通过干预差异表达miRNA调控Hh信号通路而抗肝纤维化。
Objective To explore the regulatory effect of Cigu Xiaozhi pill-containing serum on the differentially expressed miRNAs related to the Hh signaling pathway.Methods Using bioinformatics technology to screen different concentrations of Cigu Xiaozhi pills in the serum intervention group for differential expression of miRNAs,perform GO and KEGG enrichment analysis to screen out the Hh signaling pathway related miRNAs,and use Q-PCR technology to verify.Results Bioinformatics screened the target genes of differential miRNAs,and the results showed:that compared with the normal group,the 2%drug-containing serum group screened out 63 differential miRNAs,34 genes were up-regulated and 29 genes were down-regulated;the 4%drug-containing serum group screened out 72 differential miRNAs,37 genes were up-regulated and 35 genes were down-regulated;the 6%drug-containing serum group screened out 82 differential miRNAs,41 genes were up-regulated and 41 genes were down-regulated.Through GO and KEGG enrichment analysis,9 significantly different miRNAs in the Hh signaling pathway were found:miR-149-5p,miR-199a-5p,miR-210-3p,miR-222-3p,miR-27b-3p,miR-34a-5p,miR-378a-3p,miR-497-5p,miR-671-5p.By consulting related literature,three target genes miR-378a-3p,miR-199a-5p,miR-34a-5p were finally determined for Q-PCR verification.The results showed that compared with the normal control group,the expression of the miR-378a-3p gene was up-regulated(P<0.01),and the gene expression of miR-34a-5p and miR-199a-5p was down-regulated in the serum treatment group with different concentrations of Cigu Xiaozhi Pills(P<0.01).Conclusion The Chinese medicine Cigu Xiaozhi Pills containing serum can cause down-regulation of miR-199a-5p and miR-34a-5p gene expression and up-regulation of miR-378a-3p gene expression in LX2 cells;Anti-hepatic fibrosis may be achieved by interfering with differentially expressed miRNAs that regulate the Hh signaling pathway.
