摘要:目的观察参附注射液对大鼠肝缺血再灌注胆道损伤后的影响及可能的保护机制。方法按照体重将SD雄性大鼠随机分为3组:假手术组、模型组和实验组,每组10只。造模前,实验组大鼠腹腔注射参附注射液10 mL·kg-1;假手术组和模型组给予相同剂量的0.9%NaCl,1次/天,连续7 d。建立70%的大鼠肝缺血再灌注损伤模型。缺血1 h再灌注4 h后,用酶联免疫吸附法检测血浆肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)和白细胞介素10(IL-10)含量,用免疫组化法检测胆道组织核转录因子(Nrf2)和血红素加氧酶(HO-1)的表达水平。结果假手术组、模型组和实验组的血浆TNF-α的含量分别为(20.03±1.84),(79.16±6.48)和(52.01±6.54)pg·mL-1;上述这3组的血浆IL-1β含量分别为(12.41±3.62),(158.42±6.89)和(87.49±4.74)pg·mL-1;上述这3组的血浆IL-10含量分别为(17.56±1.27),(35.98±7.21)和(120.11±0.75)pg·mL-1;上述这3组的胆道组织Nrf2的表达水平(灰度值)分别为6.98±0.73,23.85±1.94和40.98±4.31;上述这3组的HO-1的表达水平(灰度值)分别为6.01±0.14,29.36±0.12和43.62±5.34。模型组与假手术组比较,上述指标的差异均有统计学意义(均P<0.01);实验组与模型组比较,上述指标的差异均有统计学意义(P<0.05,P<0.01)。结论参附注射液具有显著改善大鼠肝缺血再灌注后胆道损伤的作用,其机制可能与上调Nrf2和HO-1蛋白表达、降低促炎因子水平和促进抗炎因子的释放有关。
Objective To observe the effect of Shenfu Injection on bile duct injury in rats after hepatic ischemia reperfusion and its porentional mechanism.Methods The rats were divided into three groups:sham operation group,model group and expermental group.The experimental group was preoperatively administered,Shenfu Injection 10 mL·kg-1was intraperitoneal injection each day,for 7 days.The sham operation group and model group were given the same volume of 0.9%sodium chloride injection.Rat model of hepatic ischemia reperfusion injury was established.Contents of tumor necrosis factorα(TNF-α),interleukin-1β(IL-1β)and IL-10 in rat blood plasma were detected by enzyme linked immunosorbent assay.Expression level of nuclear factor(Nrf2),heme oxygenase-1(HO-1)in biliary tissue were detected byimmunohistochemistry.Results The TNF-αcontent of plasma in sham operation group,model group and expermental group were respectively(20.03±1.84),(79.16±6.48),(52.01±6.54)pg·mL-1;the IL-1βcontent in the above three groups were respectively(12.41±3.62),(158.42±6.89),(87.49±4.74)pg·mL-1;the IL-10 content in the above three groups were respectively 17.56±1.27,35.98±7.21,120.11±0.75 pg·mL-1;the Nrf2 expression level(grey value)in the above three groups were respectively 6.98±0.73,23.85±1.94,40.98±4.31;the HO-1 expression level(grey value)in the above three groups were respectively 6.01±0.14,29.36±0.12,43.62±5.34.Significant difference was found between sham group and model group(all P<0.01),and significant difference was also found between the three experimental groups and model group(P<0.05,P<0.01).Conclusion Shenfu Injection had significantly improved the effect of bile duct injury in rats after hepatic ischemia reperfusion,and its mechanism may be related to the up-regulation of Nrf2,HO-1 protein,the decrease of the level of proinflammatory factors and the promotion of the release of anti-inflammatory factors.
