摘要:目的制备牛血清白蛋白(BSA)纳米载药粒子(DOX@BSA/HA),并研究其释放性能。方法以BSA为纳米载体,负载盐酸阿霉素(DOX·HCl),在酸性条件下,通过吸附作用将海藻酸钠(HA)包覆于BSA表面,制备DOX@BSA/HA,并研究其释放性能。结果动态光散射(DLS)、透射电镜(TEM)和圆二色谱(CD)的结果表明,DOX@BSA/HA具有良好的分散性,直径约130~160 nm,在不同条件下均有较好的稳定性。DOX@BSA/HA在pH=7.4时具有良好的释放性能,缓释时间可达72 h以上。结论 DOX@BSA/HA是一种优良的纳米载药粒子,可有效延长DOX·HCl的体内循环时间,为DOX·HCl的临床应用提供一定的理论依据。
Objective To prepare bovine serum albumin (BSA) nano drug-loaded particles (DOX@ BSA/ HA) and study its release performance. Methods BSA was used as nanocarrier and doxorubicin hydrochloride (DOX · HC1) was loaded. Under acidic conditions, sodium alginate (HA) was coated on the surface of BSA to pre- pare DOX@ BSA/HA by adsorption, and its drug release performance was studied.Results The results of dynamic light scattering (DLS) ,transmission electron microscopy (TEM) and circular dichroism (CD) showed that DOX@ BSA/HA has good dispersibility and its diameter is about 130 to 160 nm,which is relatively stable under different conditions. DOX@ BSA/HA showed good release properties at pH = 7.4, and the sustained release time can be up to 72 h. Conclusion DOX@ BSA/HA is an excellent nano drug-loaded particle, which can effectively improve the circulation time of DOX · HCl in vivo. The research can provide a theoretical basis for the clinical application of DOX ·HCl.
