摘要:目的:观察菖蒲郁金汤对抽动秽语综合征(TS)模型大鼠纹状体铁代谢关键蛋白和多巴胺(DA)含量的影响。方法:将48只雄性SD大鼠随机分为空白组12只,造模组36只,用亚氨基二丙腈造模。完成后将造模组随机分为模型组、西药组、中药组,每组12只。西药组予硫必利悬浊液,中药组予菖蒲郁金汤药液。在造模后、干预后对大鼠进行行为学观察;用RT-PCR法检测酪氨酸羟化酶(TH)、二价阳离子转运蛋白1(DMT1)、膜铁转运蛋白1(FPN1)、铁调素(Hepcidin)mRNA的表达;免疫组织化学法检测DMT1、FPN1、Hepcidin蛋白表达情况;免疫荧光法检测TH表达;普鲁士染色检测纹状体神经细胞铁表达;ELISA法检测纹状体DA及高香草酸(HVA)表达。结果:造模后各组行为学评分均>2分;造模后及干预后,与空白组比较,模型组FPN1表达、铁、TH、DA及HVA显著降低(P<0.05),DMT1、Hepcidin表达显著增加(P<0.05);中药组FPN1、铁、TH、DA及HVA较模型组显著增加(P<0.05),DMT1、Hepcidin显著降低(P<0.05);中药组行为学评分低于模型组及西药组(P<0.05)。结论:菖蒲郁金汤可能通过调节纹状体中铁代谢关键蛋白进而调节DA含量发挥抗抽动作用。
Objective:To observe the effects of Changpu Yujin Decoction on the contents of key proteins of iron metabolism and dopamine(DA)in striatum of Tourette's syndrome(TS)rats.Methods:A total of 48 male SD rats were randomly divided into blank group(n=12)and model group(n=36),the model was made by using IDPN.After completion,the model group was randomly divided into model group,western medicine group and traditional Chinese medicine group,12 rats in each group.The western medicine group was given thiapride suspension,and the traditional Chinese medicine group was given Changpu Yujin Decoction.Behavioral observation was carried out after model building and intervention.The mRNA expression of TH,DMT1,FPNI and Hepcidin were detected by RT-PCR.The expressions of DMT1,FPN1 and Hepcidin were detected by IHC.TH expression was detected by immunofluorescence assay.Perls was used to detect the expression of iron in striatal nerve cells.The expression of DA and HVA in striatum was detected by ELISA.Results:After modeling,the behavioral scores of each group were>2.In model group,the expression of FPN1,Fe,TH,DA and HVA decreased than blank group after model building and intervention(P<0.05),while the expression of DMT1 and Hepcidin increased(P<0.05).Compared with the model group,the FPN1,iron,TH,DA and HVA in the traditional Chinese medicine group were increased(P<0.05),while the DMT1 and Hepcidin were decreased(P<0.05),and the behavioral score in the traditional Chinese medicine group was lower than that in the model group and the western medicine group(P<0.05).Conclusion:Changpu Yujin Decoction may play an anti-twitch effect by regulating the key protein of iron metabolism in striatum and then regulating the content of DA.
