摘要:目的研究四神丸对溃疡性结肠炎(UC)大鼠血清白细胞介素-2(IL-2)、肿瘤坏死因子-α(TNF-α)和细胞信号转导抑制因子3(SOCS3)表达及结肠组织病理形态的影响。方法将SPF级Wistar大鼠随机分为空白组、模型组、美沙拉嗪组及四神丸组4组,采用复合方法复制脾肾阳虚型UC大鼠模型。各组大鼠以相关药物及生理盐水灌胃21 d后处死,取大鼠结肠,肉眼观察结肠黏膜大体形态损伤并进行评分;分离各组大鼠血清,采用ELISA法检测血清IL-2,TNF-α,SOCS3的表达水平。结果与空白组比较,模型组大鼠结肠黏膜大体形态损伤评分及血清IL-2,TNF-α水平显著升高,血清SOCS3水平显著降低,差异均有统计意义(P<0.05);与模型组比较,美沙拉嗪组与四神丸组大鼠结肠黏膜大体形态损伤评分及血清TNF-α水平显著降低,血清SOCS3水平显著升高,差异均有统计意义(P<0.05)。结论四神丸可能通过调节血清TNF-α,S OCS3的水平来调控肠上皮免疫系统,从而达到治疗UC的目的。
Objective To explore the effects of Sishen Wan on the expression of serum interleukin 2 (IL-2), tumor necrosis factor-α pathological forms of colonic tissue in model (TNF-α) and suppressor of cytokine signaling rats with ulcerative colitis (UC). Methods hypothalamus cytokine 3 ( SOCS3 ) as well as SPF Wistar rats were randomly divided into control group, model group, mesalazine group and Sishen Wan group. UC rat models were re- produced by complex method. All rats were given drugs and saline by garage for 21 days. Then microscopic da- mage on colonic mucosa was observed. The levels of IL-2, TNF-α and SOCS3 in serum were detected by ELISA. Results Compared with control group, the macroscopic damage scores, the levels of serum IL-2 and TNF-α in model group increased ,while SOCS3 decreased obviously( P〈0.05 ) ; compared with model group ,the macroscopic damage scores and the levels of serum TNF-α decreased, while SOCS3 increased in mesalazine group and Sishen Wan Group. The differences were all significant (P〈0.05). Conclusion Sishen Wan can cure UC by adjusting the levels of TNF-α and SOCS3 factors to regulate and control the immune system of intestinal epithelium.
